The Experimental Study For The Role Of Heat Shock Protein70 In The Surgical Trauma

Objective:To explore the role of heat shock protein70 in the surgical trauma and homeostasis.The level of HSP70 in lymphocytes frome rats which have performed varying degrees of surgical trauma were detected and the relationship between HSP70 and surgical trauma/homeostasis was researched.Methods:1.To set up animal models:The healthy male Sprague-Dawley (SD) rats were randomly divided into four groups:(1) control group(G0 group): rats were performed without any surgical treatment;(2) experimental groups were:â‘ mild trauma group(G1 group):rats were performed the hepatic left lateral lobectomy;â‘¡moderate trauma group(G2 group):rats were performed the hepatic left lateral and right lobectomy;â‘¢severe trauma group(G3 group): rats were performed the hepatic left lateral,right and nipple right lobectomy. Each group included 20 rats which were reared in the same environment.2.To detect indicators:Taking blood samples separately in each group at 12h of preoperative,2h,12h,24h and 48h of postoperative.The HSP70 in lymphocytes were detected by using flow cytometry.The PH,HCO₃^-,BE values,K⁺,Na⁺ and Ca²⁺ were detected by STAT and fully automatic biochemical machine.The sata was analyzed by statistical software SPSS13.0.Results:(1) Successfully established model of varying degrees surgical trauma by SD rat hepatic lobectomy.The mild traumatic group survival rate was 97.5%;the moderate injury group survival rate was 95.0%;the severe injury group survival rate was 91.3%.The difference between three groups was not significant(P＞0.05).(2) HSP70 levels in lymphocytes increased with time after surgical trauma, especially reaching the peak at 12h(P＜0.05),and then decreased slowly until it's higher than the normal level at 48h;The HSP70 levels increased with varing degree of trauma(P＜0.05).It's prompt that HSP70 can be used as one of the indicators for the extent of surgical trauma.(3) The K⁺ were rapidly declined to the lowest level at 2h after trauma in three groups,but then it gradually increased to normal levels.The K⁺ was significantly decreased at 2h after surgery with the degree of trauma(P＜0.05), the others have no significant difference(P＞0.05).For Na⁺ and osmotic pressure in three groups,it was gradually decline to the lowest level at 12h,then increased to preoperative levels;The Na⁺ is significantly decreased at 12h after surgery with the degree of trauma(P＜0.05),the others have no significant changed(P＞>0.05).PH is reduced more obvious at 2h after surgery in severe trauma group,and it significant differences between other groups(P＜0.05).The PH was not obviously changed in other three groups,and no significant difference between them(P＞0.05).The BE and HCO₃^- were slightly lower than the normal range at 2h in severe trauma group,but it was no significant difference between other groups(P＞0.05).(4) For relationship between HSP70 and K⁺ in different groups,it was positive correlation after operative trauma in moderate and sever groups (P＜0.05);Also it was highly correlation at 2h and midrange correlation at 12h/24h after sugery in three different groups(P＜0.05).For relationship between HSP70 and the osmotic pressure in different groups,it was negative correlation after operative trauma in three groups.It was highly correlation at 12h/24h after sugery in three different groups(P＜0.05).Conclusion:1.The HSP70 levels in lymphocytes increased with varing degree of trauma.It's prompted that HSP70 can be used as one of the indicators for the extent of surgical trauma.2.The internal environment can be recovered at post operation by HSP70 levels.It's prompted that HSP70 plays an important role in homeostasis.3.The relationship between HSP70 and internal environment was negativeely correlated with the increasing degree of surgical trauma process.It's prompted that HSP70 and homeostasis were accompanied by various degree of trauma.We can presume that HSP70 maybe do as an early warning indicator of homeostasis,but it must be clear by further study.
