Background Cyclins are a family of proteins implicated in the induction and control of mitosis.They are characterized by their accumulation in the interphase and dramatic destruction at the end of each cell cycle via the ubiquitin pathway.The cyclins have been showed to involve in cell proliferation and hypertrophy,apoptosis and tumorigenesis.CyclinⅠhas been cloned and sequenced from an equalized cDNA library derived from human forebrain cortex.It contains a typical cyclin box near the N-terminus and a PEST-rich sequence near the C-terminus,and shows the highest sequence similarity in the cyclin box to cyclin G and E.CyclinⅠmRNA is expressed at high levels in post-mitotic tissues,including skeletal muscle,heart,and brain, and is expressed constantly during cell cycle progression,and thus functions independently of the cell cycle control.But the function of this gene isn't fully discovered.Its physiologycal rol in spermatogenesis and/or human sperm cell function,anti apoptosisi and tumor angiogenesis have been reported recently.In our previous experiments,cyclinⅠexpression in co-cultured fibroblast cells with epithelial cells showed lower than that in non-cocultured fibroblast cells, thus it maybe have some important role in the prostate.For further study the function of cyclinⅠ,it becomes the initial mission to know the expression state of cyclinⅠin normal prostate tissue,BPH tissue and prostate cancer.Objective To investigate the expression of cyclinⅠin different prostate tissue(normal prostate,BPH and prostate carcinoma) to prelimijarily research whether cyclinⅠis differentially expressed and its potential significance.Methods Using the immunohistochemical assay to detect tissue microarray slides composed of 24 normal prostate,61 BPH and 38 prostate cancer tissue.Results Immunohistochemistry utilizing cyclinⅠantibody showed that there was significant difference of the cyclinⅠexpression among the normal prostate,BPH and prostate carcinoma tissues(P=0.000).The positive expression of cyclinⅠin prostate carcinoma was significant lower than that in normal prostate tissues(P<0.01)and in BPH tissues(P<0.001).Conclusions In this study,we successfully to accomplish the Immunohistochemistry experiments utilizing anti- cyclinⅠpolyclonal antibody showed that cyclinⅠexpressed in prostate epithelial and fibroblast cells.The expression of cyclinⅠwas significantly lower in prostate cancers than that in normal prostate tissues and BPH.But the role of anti apoptosisi and tumor angiogenesis that cyclinⅠplays in the prostate need more further investigate.
