Objective To detect the effective of the azithromycin,acetyl spriarnycin,artesunat and combined medicine on Toxoplasma gondii in vitro and in vivo. To search for the active drug and approach of healing toxoplasmosis, providing sciencic instruction for clinic rational use of drug.Methods In vitro tests:To obtain the hydrops abdominis suspension of toxoplasma gondii RH strain. In germ free cell culture plate,the tachyzoites of Toxoplasma and the different concentration of drugs were added, NS act as the negative control group.The different concentration of Azithromycin,Acetyl spri- arnycin,Artesunat and combined medicine are experiment groups. Then the act- ivities of the drugs against the Toxoplasma gondii were assayed by viewing cell counting and MTT staining, and then calculating fractional inhibition, drawing the medicine dose effect curve.In vivo tests : To set up acute toxoplasmosis animal model of mice . Two hours after infection,the mice were divided into Azithromycin group(A),Acetyl spriarnycin group(B),Artesunat group(C),Azithromycin and Artesunat combi-ned group(D),Azithromycin and Acetyl spriarnycin combined group(E),Artes-unat and Acetyl spriarnycin combined group (F),at the same time, seting up untreatment model as control group. Initial dose redup, recording experiment mice,s food and drink,activity,incidence,death time,detecting the morphous and density test death Irrigation of peritoneal cavity of toxoplasma gondii under microscope. The mice were defeceted 30 consecutive days. To compare the sur- vival days , drawing survival curve.Results In vitro tests:(1) morph-observation: after 72h drug action, re- viewing the marked change figure every experimental group (Azithromycin,ac- etyl spira-mycin,Artesunate,and several drugs combination) under optical mi- croscope,but the blank control group has no marked change.In vivo tests : after 24h drug action,when the concentration of Azithro- mycin are 0.2ug/mL and 200ug/mL, fractional inhibition are 35.7﹪and 68.8﹪. But when the density of acetyl spiramycin were 0.2ug/mL and 200ug/mL, fract- ional inhibition are 2.56﹪and 8.25﹪. The density of Artesunate were 2.5ug/mL and 2500ug/mL, fractional inhibition are 7.7﹪and 51.0﹪. The density of the combined group(acetyl spiramycin+ Artesunate)are 4ug/ml+5ug/m and 4000ug/ ml+5000ug/m, fractional inhibition are7.3﹪and 46.3﹪.Dose response curve to show that several drugs all have positive correlation the dose-effect relation.In vitro tests:after 3 days action, experimental group and control group all have no conspicuous Abnormal appearance. When the the mice starting occu- rrence activity weaken,chilly,lethargy,hair disperative replication for control group(G)and acetyl spiramycin group(B), acetyl spiramycin and Artesunate combind treatment group(F) and when the fiveth day the mice starting occurren- ce death,When the sixth day, Azithromycin group and acetyl spiramycin comb- ind treatment group(E),Artesunate group (C)the mice starting occurrence death. When the seventh day, Azithromycin group (A),Azithromycin and Artesunate combind treatment group (D)the mice starting occurrence death.But control gro- up all are death within seven days, acetyl spiramycin treatment group could sur- vive for eight days.Other group starting occurrence light symptom,the to exist or last for 15 days,to make use of Log-rank statistical analysis,to compare the experimental group and control group,have significant disparity,has statistical significance(p<0.05).Conclusions The azithromycin,acetyl spriarnycin,artesunat and combined medicine could inhibit multiplition of T.gondii. At the common dosage,the kiling efect of the azithromycin on the tachyzoites of Toxoplasma gondii is better than other drugs and shows a positive correlation with the dosages in vitro.Administering could elevate the survival days of infected experimental animal.Under conventional therapy of drug concentration, the kill effective of toxoplasma gondii by turns are Azithromycin,Azithromycin and Artesunate combined treatment group,Artesunate,Azithromycin and Artesunate combined treatment group,Artesunate and acetyl spriarnycin combined treatment group,acetyl spriarnycin.Azithromycin,Azithromycin and Artesunate combined treatment group could become fairly better adoption for clinic treatment.Exploring and erecting stabile the MTT drug sensitive empirical method, screening and evaluating the drugs anti- T.gondii , providing Ideal investigate platform.
