Hepatocellular carcinoma (HCC) is the fifth most common cancer in the world. About 18.8% of total fatalities yearly caused by malignant tumor are HCC in China, and HCC is the most common cancer in Guangxi .However, the molecular mechanisms of hepatocarcinogenesis are remaining largely unknown. Currently, the research of HCC is concentrated on related gene cloning, cellular signal transformation pathway and regulation of cell cycle. Wnt signaling pathway is hotspot to study recently. Many evidences indicate that Wnt signaling pathway may play a key role in the hepatocarcinogenesis. However, the exact mechanism of Wnt signaling pathway in hepatocarcinogenesis is still uncertain.As a oncogene protein, Twist is considered to be closely related encoding inhibitor of apoptosis protein, tumor cell apoptosis resently. Twist is associated with invasion and metastasis of hepatocellular carcinoma.Objective: To investigate the relationship between Twist gene and the Wnt signaling pathway in SMMC7721 Hepatoma cell line .Methods: Wnt signal transduction pathway in SMMC7721 Hepatoma cell line was effcted by lithium chloride, a activator of Wnt pathway. RT-PCR and fluorescence quantitative PCR were detectedβ-Catenin and Twist gene expression.Results: After using lithium chloride,β-Catenin gene expression in SMMC7721cell line is higher than pre-activation (P<0.05). Meanwhile, Twist gene overexpression was identified by RT-PCR and fluorescence quantitative PCR,respectively.Conclusion: The relationship between Twist gene and Wnt signaling pathway in hepatocellular carcinoma cell was associated .
