Malondialdehyde(MDA) is one of the representative toxic carbonyl substances being produced in the progress of lipid peroxidation and non-enzymatic glycosylation.It may cause the carbonyl-amino cross-linking and lead to the aging-associated accumulation of irreversible changes,such as crosslinked collagen and lipofuscin formation in the post-mitotic cells.This paper is to investigate effects of MDA and serotonin(5-hydroxytryptamine,5-HT) over erythrocytes on the basis of the carbonyl stress hypothesis of aging developed from our laboratory.This study includes two parts:Part one was aimed to studied the effects of 5-HT on erythrocytes in relation with carbonyl-stress.Three experiments were included in this part describe as follows:Firstly,measuring blood viscosity under different treatments.The results indicated that MDA induced carbonyl stress in blood.And 5-HT could intensify this tendency on erythrocyte under carbonyl stress. Secondly,measuring osmotic fragility of erythrocyte.The results showed that osmotic fragility of erythrocyte treated with MDA was larger than blank-treatment.The values of three serotonin-treats with different concentrations were higher than the control.Lastly,observing morphological alterations of erythrocytes by fluorescence microscopy. Serotonin was found to intensify aggregation,alteration of cell morphological changes after MDA treatment in the suspension.And it seemed that some substances in the plasma could protect the erythrocytes.Part two was to explore the effects of de-carbonylation function of lysine on the exhaustive exercise of rats.After adaptive running for 3-d training exercise,32 rats were divided into four groups stochastically(one control group and three experimental exercise groups).While the three exercise groups were fed with different chemicals,the control group was fed with saline before study.The experimental rat groups were carried through exhausting exercises according to Bedford-model.Following the exhausting exercises,all the rats were sacrificed immediately and blood plasma was obtained by eyeball.Then the density of lactic acid,MDA and lysine in the serum were measured and analyzed respectively.The results suggested that lysine can prevent remarkably the MDA induced fatigue situation of rats during exhausting exercises.
