The distrbution of Livin had a high selectivity,tissue-specific expression in human fetal tissues and nearly all solid malignancies.Following the research, Livin became a new focus in inhibit apoptosis and induce cell multiplication. Bcl-2(B-cell lymphoma/leukemia-2)which is one kind of oncogenes separated from the cell of follicular lymphoma. Bcl-2 antagonizes the process of apoptosis. Bcl-2 expressed majority normal tissue and tumor of people and concerned with differentiation,drug resistance and prognosis of tumor.The aim of this study was to analyze the infouence of Livin and Bcl-2 to esophageal squamous carcinoma occurrence and development by examining the expressivity of Livin and Bcl-2 in the tissue of esophageal squamous carcinoma and provide clinical research of esophageal carcinoma with scientific theory.Methods: Applying flow cytometry was used to detect the levels of Livin and Bcl-2 in esophageal carcinoma. The esophageal squamous cancer tissues and esophageal pericarcinoma tissues specimens were all from the esophageal disease patients therapied in the Department of Thoracic Surgery of the Forth Hospital of Hebei Medical University from 2008.8 to 2008.10, of which 50 cases belonged to esophageal squamous carcinoma tissue, among them male 32 cases, female 18 cases, the age from 43 to 72 years old. According to the standard of TNM staging(UICC, 2002), 6 cases belong to T1 stage, 6 cases belong to T2 stage, 29 cases belong to T3 stage, 28 cases belong to N0 stage, 22 cases belong to N1 stage. The 50 cases pericarcinoma tissue was taken for comparison. Specimens were incised into 1 to 2 pieces which about 0.5cm×0.5cm×0.5cm. Then one by one in order carry on filtering, centrifuging, labeling, and then the single-cell suspension liquid was detected by adopted to Folw cytometry. PBS solution was taken to be negative control and FITC solution to be positive control. Fluorescence intensity(mean±standard deviation) represent the testing result. The operations of the end are used SPSS13.0 statistical software to analyze examination result, included t test,analysis of variance, Pearson correlation analysis, etc. Results:1 The expression of Livin in the different tissue: The average fluorescence intensity of Livin in esophageal squamous carcinoma was 197.87±50.13,the average fluorescence intensity of Livin in esophageal pericarcinoma tissues was 78.56±39.81. The difference of Livin expression between esophageal carcinoma tissues and esophageal pericarcinoma tissues was significant(P<0.05).2 The expression of Bcl-2 in the different tissue: The average fluorescence intensity of Bcl-2 in esophageal squamous carcinoma was 322.93±54.77,the average fluorescence intensity of Bcl-2 in esophageal pericarcinoma tissues was 164.49±47.46. The difference of Bcl-2 expression between esophageal carcinoma tissues and esophageal pericarcinoma tissues was significant(P<0.05).3 The relationship between the expression of Livin and Bcl-2 in esophageal squamous carcinoma and clinocopatholonical characteristics: No correlation was observed between expression intensity of Livin and Bcl-2 and some clinocopatholonical characteristics(age, gender, histodifferentiation; There was association between the expression of Livin or Bcl-2 in esophageal squamous carcinoma or infiltration, the difference was significant (P<0.05); There was association between the expression of Livin or Bcl-2 in esophageal squamous carcinoma or lymph node metastasi, the difference was significant (P<0.05). 4 The relationship between the expression of Livin and Bcl-2 in esophageal squamous carcinoma and TNM staging(UICC): in esophageal squamous carcinoma the average fluorescence intensity of Livin at stageâ… was 108.93±18.07,at stageâ…¡a was 160.96±36.70,at stageâ…¡b was 194.31±32.03,at stageâ…¢241.91±22.58.The difference of Livin expression between different stage was significant(P<0.05). The average fluorescence intensity of Livin at stageâ… was 227.13±21.34,at stageâ…¡a was 271.97±23.93,at stageâ…¡b was 322.63±36.25,at stageâ…¢369.37±23.37.The difference of Livin expression between different stage was significant(P<0.05).5 The correlation between the expression of Livin and Bcl-2: the average fluorescence intensity expression of Livin in esophageal squamous carcinoma was 197.87±50.13, the average fluorescence intensity expression of Bcl-2 in esophageal squamous carcinoma is 322.93±54.77. There was a positive correlation between the expression of Livin and Bcl-2 in esophageal squamous carcinoma (r=0.856 , P<0.01). The average fluorescence intensity expression of Livin in esophageal pericarcinoma tissues was 78.56±39.81; The average fluorescence intensity expression of Bcl-2 in esophageal pericarcinoma tissues was 164.49±47.46. There was no correlation between the expression of Livin and Bcl-2 in esophageal pericarcinoma tissues (r=0.081,P>0.05).Conclusion:1 The expression of Livin and Bcl-2 in esophageal squamous carcinoma tissue was obviously higher than them in esophageal pericarcinoma tissue.2 No correlation was observed between expression intensity of Livin and Bcl-2 and the patients age, gender and histodifferentiation of esophageal squamous cancer. There was association between the expression of Livin or Bcl-2 in esophageal squamous carcinoma and lymph node metastasi or infiltration. The expression of Livin or Bcl-2 which have lymph node metastasi or the deeper infiltration is higher.3 There was association between the expression of Livin or Bcl-2 in esophageal squamous carcinoma and TNM staging. Following the stage increase the expression of Livin or Bcl-2 is higher.4 There were positive correlation between Livin and Bcl-2 in carcinoma tissue and pericarcinoma tissue. When Livin presented high expression, Bcl-2 presented presented high expression too.
