Objective: The pupose of the stndy was to detect the expression of FADD(Fas-associated death domain) protein and mRNA in LSCC (Laryngeal squamous cell carcinoma,LSCC), to find the assocication between expression of FADD and clinical pathological differentiation stage andlymph nodes status of LSCC and to evaluate the possible effect and mechanism of FADD on the development of LSCC,to provide for a new ideas for tageted therapy in LSCC.Method:1 The expression of FADD protein were detected by Immunohistochemistry (IHC) method in 30 cases of LSCC, 20 cases of papilloma of larynx and 20 cases of normal adjacent mucosa.2 The expression of FADDmRNA were detected by reverse transcription polymerase chain reaction (RT-PCR) method in 10 fresh cases of LSCC and 10 cases of normal adjacent mucosa. SPSS13.0 systerm was used to perform the statistical analysis, have statistical significance (P<0.05).Results:1 The results of immunohistochemistry: The positive expression rate of FADD protein in LSCC group, papilloma of larynx group andnormal adjacent mucosa group were 76.7%,90.0%,95.0%,the positive expression rate have statistical significance in total(P<0.05).The positive expression rate in LSCC lower than the normal adjacent mucosa group, have statistical significance between them(P<0.05); The positive expression rate in LSCC was lower than papilloma of larynx group,but with no statistical significance(P>0.05); Between the papilloma of larynx group and the normal adjacent mucosa group,with statistical significance (P<0.05). The positive expression rate of FADD protein were obtained to relate cinical stage with the degree of pathological differentiation,that is: the stage of I-II group in LSCC were 95.2%, the stage of III-IV were 50.0%, with statistical significance (P<0.05); In LSCC of high differentiated were 94.4%, higher than middle-poorly differentiated were 25.0%, with statistical significance (P<0.05).But no related to sexuality ,age ,clinical classification and the lymph node metastasis,have no statistical significance was obtained (P<0.05).2 The results of PCR: The expression of FADDmRNA were 0.485±0.141 in LSCC group much lower than in normal adjacent mucosa group which is 0.892±0.265,have statistical significance (P<0.05). The expression of FADDmRNA were obtained to relate cinical stage with the degree of pathological differentiation,that is: the stage of I-II group in LSCC were 0.593±0.024, the stage of III-IV were 0.376±0.117, with statistical significance (P<0.05); In LSCC of high differentiated were 0.568±0.053, higher than middle-poorly differentiated were 0.318±0.081, with statistical significance (P<0.05). But no related to clinical classification ,have no statistical significance was obtained (P<0.05).Conclusion:1 The positive expression rate of FADD in groups of normal adjacent mucosa and papilloma of larynx much higher than the group of LSCC.Which implys that FADD plays an important part in the occurrence and development of carcinoma of larynx.2 Later clinical stages and differentiation FADD positive expression rate of the much poorer lower, to detect the expression rate of FADD may provide guidance for clinical prognosis.3 As a key factor in the induction of apoptosis, FADD plays an important role in the apoptosis signal transduction pathway,inducing apoptosis,whether it will be a means of target treat tumor still to be further explored.
