Objective: psoriasis is a common chronic inflamed disease of the skin with strong genetic and environmental risk factors which has proliferation acceleration of epidermis and parakeratosis. At present,it is not completely that we know the cause of disease and pathogenesises of the psoriasis.But the immunity function abnormality is an important factor. T lymphocytes are thought to be the central in its pathogenesis. More and more studies reported that there are regulatory T cell confused in the peripheral blood of the psoriasis patient. The blance between regulatory T cells and effector T cells is important for normal immune function. utill recently there have been more studies on CD4~+CD25~+regulatory T cells and more and more sports on the suppressing mechanism and clinical significance.Altough there have been some substantial advances in the understanding of the mechanism for their development including the site where they are generated, the cells and the signal molecules that are required for their developments. The understanding of these,At present,little is known about the differentiation of the CD4~+CD25~+regulatory T cells .Which originated from the thymus, play an important role in the maintenance of self immune tolerance. As an important subset of the regulatory cells,the CD4~+CD25~+Treg cells have the features of hyporesponsive and anergic to antigen stimulation. Recently, more and more researches have indicated that CD4~+CD25~+ Treg cells play an important role in the suppressing the activation of self-reactive T cells and maintaining the immune tolerance in the autoimmunity mainly by secreting immunosuppressive cytokines such as TGF-βand IL-10, Furthermore, their number absence or functional deficiency may lead to some autoimmune diseases. Regulatory CD4~+CD25~+T cells play a key role in the negative regulation of the immune system, which can inhibit inappropriate self-reactive T cells, as well as limit the range, extent and lasting time of immune responses, so as to inhibit the proliferation of CD4+ and CD8+ effector T cells. Many recent researches have indicated that CD4~+CD25~+ Treg cells play an important role in the certain skin tumors and skin diseases,such as psoriasis and systemic lupus erythematosus. Functional CD4~+CD25~+Treg cells play an important role in the maintenance of homeostasis, anti-infection, induction of transplantation-tolerance.In this study, we measured peripheral blood levels of CD4~+CD25~+regulatory T cells the in psoriasis vulgaris patients and the control group to observe the function of the CD4~+CD25~+regulatory T cells in pathogenesis of the psoriasis vulgaris.It can reveal the pathogenesis of the psoriasis and provide us another new way for the therapy of the psoriasis. Methods: The case group involved thirty-five psoriasis vulgaris patients who were selected in out-patient clinic in the second hospital of Hebei Medical University from April, 2007 sepetember to June, 2008. they all conform to the psoriasis vulgaris diagonsis standard. Grouping: 1. progressive psoriasis group,19 patients inclued 2. static psoriasis group,16 patients inclued. We excluded patients with autoimmunity diseases such as pemphigus, erythema lupus, dermatomyositis,rheumatic arthritis, et al, patients with other severe systemic diseases, with tumor .All the cases involved had not taken any drugs that could affect function for three months such as corticosteroids and immuno suppressive agent, et al. The control group involved twenty healthy people without family history and immunological diseases. With respect to thre age,the gender, there was not significant difference between the two groups by analysis of t-test. two milliliters blood samples from healthy controls and patients after PASI score were collected and separated. Peripheral blood levels of the CD4~+CD25~+regulatory T cells were measured by Flow cytometry according to directions in the test kit. Statistical analysis was performed using one-way ANOVA and pearson correlation, A P value of less than 0.05 was considered as significant.Results: 1.The levels of CD4~+CD25~+regulatory T cells in the Peripheral blood of Patients with progressive psoriasis were significantly lowerer than that of the normal controls and whom with static psoriatic patient(P<0.05); 2.In psoriasis vulgaris patients,the percentages of CD4~+CD25~+Regulatory T cells among T cells from the peripheralblood was not correlated with PASI score.. (r=0.151,P>0.05).In progressive psoriasis vulgaris patients, the percentages of CD4~+CD25~+Regulatory T cells among T cells from the peripheral blood was not correlated with PASI score.. (P>0.05).In static psoriasis vulgaris patients, the percentages of CD4~+CD25~+Regulatory T cells among T cells from the peripheral blood was not correlated with PASI score.. (P>0.05)3. In psoriasis vulgaris patients, the percentages of CD4~+CD25~+Regulatory T cells among T cells from the peripheral blood was not correlated with duration. (r=0.209, P>0.05)。Conclusion:1.The CD4~+CD25~+Regulatory T cells maybe participate the invasion of the psoriasis,by suppressing the activation of self-reactive T cells. No statistically significance was seen in the levels of CD4~+CD25~+Regulatory T cells between normal controls and static psoriatic patients2.The psoriasis vulgaris exist immune disequilibrium3.The percentages levels of CD4~+CD25~+Regulatory T cells among T cells do not have the direct proportion with serious degree of pathogenetic condition and duration.
