At present, subunit influenza vaccine with very pure antigen component is the first-choice vaccine for children and the elderly abroad, but it can not obtained with high protection efficience. The research on liposome as vaccine adjuvants can improve the successful immunizing rate of sub-unit vaccine with a single inoculation.This article mainly study on liposomes as adjuvant of subunit influenza vaccine which has not been marketed in China.Based on the technical platform of sub-unit influenza vaccine,our research prepared large multi-lamellar liposomes by film evaporation combined freezen-drying. As observed under transmission electron microscope, the liposomal was round or oval, has a uniform virion size distribution, and has an encapsulation rate of 33.5%. As shown by results, the liposomal influenza vaccine of lower antigen amount can stimulate body to produce higher humoral immunity and a certain cellular immunity.In a word,this article optimized various key parameters of manufacture process according to requirments of scale-up,and the immunological datas indicated that liposomal vaccine had a higher protection rate and a cellular immunity different from that of conventional influenza vaccine without adjuvants. This research result provided the beneficiary information for further study on liposomal influenza vaccine, and the foundational basis for study on new generation influenza vaccine.
