ObjectiveThe chronic inflammation of airway and remodeling of airway are asthma's important feather, which are closely correlated with the function of airway smooth muscle. The newest study said , airway smooth muscle cell has the function of immunological regulation,besides the feather of construction,hyperplasy,immigration. ASMCs could generate a lot of cytokine,chemotatic factor,growth factor,cell adhesion factor and extracellular matrix by autocrine and paracrine,which plays an important role in starting and maintaining asthmatic airway inflammation and airway remodeling. The new cognitions of ASMCs's immune function, which make mechanism which disusses the regulation of ASMCs's immune function as the new focus of asthmatic pathogenesy and the new target of asthmatic curation.The in-vitro studies indicate that the inflammtation factor such as IL-1β,TNFαwhich could induce nonasthmatic ASMCs to secrete lots of immunological regulation factor such as cytokine,chemotatic factor,growth factor,cell adhesion factor and extracellular matrix. These immunological regulation factor could regulate B lymphocyte and T lymphocyte's proliferation and immune globulin secretion and to promote the recruitment and survival of inflammation cell such as eosinophile. At the other hand, it could promote ASMCs self-generation and self-differentiation and play an important role in starting and maintaining asthmatic airway inflammation and airway remodeling. But the mechanism which contols the immune fuction of asthmatic ASMCs is not understanded at present internal and abroad.Airway smooth muscle and ECM as the important ingredient which could keep airway's structure and function, the interaction of those play the important role in asthmatic airway inflammation and airway remodeling. In the development of asthmatic patho-process, the amount of ASMCs increases and the function of ASMCs alters,at the same time , the amount and constituent of ECM alter. ECM can regulate the biological behaviour of ASMCs, such as survival,proliferation,immigration and so on. But whether ECM can regulate the immune function of asthmatic ASMCs and how to regulate it, we have no idear at present. RANTES,eotaxin,TGF-β1 would participate in asthmatic airway inflammation and remodeling.At this experiment, the problem which we discuss is how ECM would regulate the expression of RANTES,eotaxin,TGF-β1 of airway smooth muscle cell in asthmatic rat.MethodsWistar male rats were randomly divided into two groups: asthmatic group (n=8) and control group (n=8). Asthmatic model was established through the OVA sensitization and atomization. We cultured the rats'airway smooth muscle cells, made them effected by FN,LA,COL-I. The expression of RANTES,eotaxin,TGF-β1's mRNA and protein were detected by real time PCR and western-blot.Results RANTES,eotaxin and TGF-β1's expressions in non-effeceted asthmatic group were significantly higher than non-effeceted control group(P<0.05); they in effeceted asthmatic group were significantly higher than non-effeceted asthmatic group(P<0.05); but they in control group were no ignificant difference(P>0.05).ConclusionECM may participate in regulating the immune function in asthmatic airway smooth muscle.
