Part one In vitro culture and identification of endothelial progenitor cells(EPCs) in peripheral blood of Wistar ratsAimsTo explore the intro culture and identification methods of endothelial progenitor cells in peripheral blood of Wistar rats.MethodsWe collected peripheral blood of Wistar rats from hearts, then isolated mononuclearcell(MNC) with density gradient centrifugation, then cultured in EBM-2 medium.We used Dil markered acLDL uptake test,FITC markered UEA combination test and immunofluorescence assays detected the expression of Dil-acLDL,FITC-UEA-1.ResultsEPCs were successfully cultured from the MNC in peripheral blood of Wistar rats, those EPCs can phagocytose acLDL an combine with UEA-1.Its phenotypes were Dil-acLDL+,FITC-UEA-1+. ConclusionEPCs were successfully cultured from the MNC in peripheral blood of Wistar rats with density gradient centrifugation.Part two Effect of endothelial progenitor cells transplantation on neovasculariza- tion and cardiac function in experimental acute myocardial infarction ratAimsTo explore the effect of endothelial progenitor cells(EPCs) on promoting neovascularization and cardiac function after transplantation in acute myocardial infarction rats.MethodsEPCs of Wistar rats were isolated and cultured. Immunofluorescence assays was used to detect the expression of Dil-acLDL+,FITC-UEA-1+. EPCs labeled with Brdu (experimental group)or EMB-2(medium control group) were implanted into the ischemic myocardium resulting from ligation of left antierior descending branch of coronary artery. Fifteen rats were set as sham-operated group. 4 weeks after transplantation, echocardiography was used to examine the effects on heart function, and specimens were taken from infarct zone for examining cell morphology and measuring capillary density. ResultsEPCs were successfully isolated and cultured and its phenotypes were Dil-acLDL+,FITC-UEA-1+. The labeled EPCs were successfully implanted into the ischemic area and differentiated into endothelial cells. The capillary density in infarcted area in experimental group increased compared with control group (P<0.01). Heart function, which shown as ejection fraction and fractional of shortening,was improved in experimental group compared with control group at 4 weeks(P<0.01). Histological study showed that transplanted neonatal cardiomyocyte were detected in all host hearts and identified by BrdU staining.ConclusionEPCs transplantation can accelerate angiogenesis in infarct zone after rat myocardial infarction and improve heart function.
