ObjectiveTo study the susceptibility of offsprings to glaucoma under the condition that CYP1B1 null female mice accepting certain doses of corticosteroids in pregnancy. MethodsWe took 30 female CYP1B1 null mice and 15 male CYP1B1 null mice. They were 9-10weeks. We divided them into 15 cages, and each cage had 2 female mouse and 1 male mice. At next day 9 o'clock, we took male mice away from each cage. We signed the female with embolus. At last forteen female mouse became pregnant. We randomly divided conceive mouse into 2 test groups (n=7). Test group 1 were injected 0.05ml/10g with 0.5% dexamethasone and Test group 2 were injected 0.1ml/10g with 0.025% dexamethasone. The process lasted for 12 days. The gestational period of conceived mouse was about 19-21 days, and the young mice were ablactated when they were 22 days old. Test group 1 owned 26 little mice, but test group 2 only 1 conceive mouse gave birth to a dead baby. The C57BL /6J mice grouped and managed were similar with CYP1B1 null mice. The control group 1 owned 18 conceive mice and 30 little mice. Control group 2 didn't own little mouse. As the result, our test studied test group 1 and control group 1 mice.We chose 20 young CYP1B1 null mice and 20 young C57BL /6J mice. After testing their transverse diameter of cornea and intraocular pressure,, we randomly divided 2 group mice into four groups, each group had 10 mice. For one guoup, the retinal morphology and structure of CYP1B1 null mice and C57BL /6J mice were studied by lightmicroscope, for the other, The apoptosis of the retinal cells was assayed by terminal deoxynucleotidyl transferase-mediated dUTP-biotin nick end labeling ( TUNEL ).ResultsThe corneal diameter of CYP1B1 null mice was 2.98±0.18mm, and that of C57BL /6J mice was 2.96±0.21mm. No significant difference of corneal diameter was detected between this two kinds of mice(P>0.05). The intraocular pressure(IOP) of CYP1B1 null mice was 19.6±2.6mmHg, while the C57BL /6J mice's was 16.4±3.0mmHg. there are significant difference of IOP between this two kinds of mice(P<0.05). In CYP1B1 null mice, the retinal nerve fiber layer(RNFL) and the retinal ganglion cells(RGCs) were more severely damaged compared with C57BL /6J mice. For the 20 eyes of CYP1B1 null mice, the apoptosis cells in the ganglion cell layer and inner nuclear layer were found in 14 eyes; for the C57BL /6J mice, only 5 eyes were detected with apoptosis cells. ConclusionThe offsprings of CYP1B1 null mice are more likely to generate glaucoma than C57BL /6J mice when their mother accepting certain doses of corticosteroids in pregnancy.
