In china,Cottomrose Hibiscus,Hibiscus L Malvaceae,is a kind of wide-spreaded vegetation resources.More and more interest is focused on the research on it in recent years,but little on its antitumor activity.Furthermore,almost all pharmacological researches were on flowers or leaves of Cottomrose Hibiscus and none on roots.In this article,original extracts from Cottomrose Hibiscus roots were made and Bacteriostasis and anti-tumor activities were observed in order to provide further evidence to develop this natural medicine.The solvent-refining was used to extract Cottomrose Hibiscus roots.The total ethanol,ethyl acetate(EtOAc),n-butanol(n-BuOH) and water extracts of Cottomrose Hibiscus roots were extracted.The Bacteriostasis activities were tested by serial dilution methods.The agar dilution method was used to determine minimum inhibitory concentration(MIC) of the different extracts from Cottomrose Hibiscus roots.Effect of different extracts from Cottomrose Hibiscus roots on growth of different cancer cell strains were observed by MTT method.Safety of the total ethanol extracts of Cottomrose Hibiscus roots was envalued by acute toxicity test.It has been found out that the different extracts from Cottomrose Hibiscus roots can suppress the growth of Staphylococcus aureus(ATCC29213),Escherichia coli(ATCC29523),Pseudomonas aeruginosa(ATCC27853),Staphylococcus epidermidis.The ethyl acetate(EtOAc) extracts of Cottomrose Hibiscus roots was most effective,and the MIC are 6.25mg·mL-1,12.5mg·mL-1,6.25mg·mL-1 and 12.5mg·mL-1,respectively.Within the concentration from 1.6 to 1000μg·mL-1,The ethyl acetate(EtOAc) extracts of Cottomrose Hibiscus roots can obviously inhibit the growth of cancer cell strains HL-60,K562,KAR,MCF-7 and AGS,which relies on dose and the IC50 are 332,93,54,128 and 500μg·mL-1,respectively.No distinct inhibition was found in CA-46 and less inhibition was observed in the same cancer cell strains of the total ethanol,n-butanol and water extracts of Cottomrose Hibiscus roots.To study the toxic reaction of Cottomrose Hibiscus roots in mice, the maximum administration dosage of the total ethyl acetate extracts from Cottomrose Hibiscus roots were administrated in mice by ig,twice per day.Acute toxic reactions were observed for 14 days.All mice survived,and no toxic reaction was observed during the period.The maximum administration dosage given to mice was 233.74g of crude drug per kilogram,equal to 111 times of daily dosage.So,ethyl acetate extracts from Cottomrose Hibiscus root has little acute toxicity and it is safe for oral administration.
