| Objective To observe the change of NO concentration and pressure in the acute ureteral obstruction model of dogs after given the drug,and explore the relationship between NO concentration and ureteral pressure changes.Method Using a balloon to incarcerate the lower ureter to simulate the model of acute ureteral obstruction. After the success of the models,divided them into 5 groups according to whether or not pre-given L-NAME(non-selective nitric oxide synthase inhibitor) and the doses: the A group,pre-given saline;the B1 group,pre-given L-NAME5mg;the B2 group, pre-given L-NAME10mg;the B3 group,pre-given L-NAME20mg;the B4 group, pre-given L-NAME30mg.After 10min,each group give isoprenaline 10ug/kg by intravenous,meanwhile measuring the pressure(UP),the NO concentration,the urine leakage from the balloon edge(UFO) of the obstruction ureter and blood pressure(MBP),heart rate(HR) of the model.Result After administration,at a certain period:compared the extent of UP,MBP,HR change,UFO,the total and peak of NO concentration between A group,B1 group and B2 group,the difference are not statistical significance(P>0.05);compared the indexes between B3 group,B4 group and A group,B1 group,B2 group,the difference are statistical significance(P<0.05), the extent of UP,MBP change,UFO,the total and peak of NO concentration in the former two grounps are obviously much smaller than the behind three groups,but the extent of HR change is higher,compared the indexes between B4 group,B3 group the difference are statistically significant(p<0.05),the extent of UP,MBP change,UFO, the total and peak of NO concentration in the former grounp are obviously much smaller than the behind group,but the extent of HR change is higher.Conclusion NO can reduce the pressure of the acute obstructive ureter,making the obstructive ureter recanalization,and when the more concentration of nitric oxide,more pressure will be reduced,and more urine will be collected.β-adrenergic receptor agonists can relax the ureteral smooth muscle and activate the NOS,increase NO releasing on the ureteral Organize,the part of function of reducing the pressure of the obstructive ureter is mediated by NO. |
