Objectve Endothelial dysfunction is identified to initiate and aggravate thevascular complications induced by high glucose.High glucose plays a central role inthe regulation of endothelial cells proliferation and apoptosis .It is studying the impactof intermitternt high glucose and constant high concentration of glucose onproliferation and apoptosis of endothelial cell. Methods Human umbilical veinendothelial cells were studied by CCK-8 to detect the rate of cell apoptosis andHoechst to detect the rate of cell proliferation . ResultsIn high glucose conditions, especially intermittent high glucose condition, cellproliferation rate were significantly lower than that in the normal concentrationconditon 5th day until the 7th day after interfered with high glucose .When the cellsin 20.5mmol / L glucose concentrations, the first five days of cell proliferation rateand the normal concentration of glucose compared to non-group (P> 0.05), the 6th dayand the 7th day, cell proliferation rate was significantly reduced (P <0.05). Andintermitten high glucose group compared to the constant group ,the cell proliferationrate at various time points were significantly reduced (P <0.05). In addition, the cellproliferation were lower in high glucose conditions than normal group.Conclusioncompared with the constant high glucose, intermittent high glucose has the morestrong damaged effects on endothelia cells.
