Objectives1.To study the mRNA expression of Cks1 and Cks2 in HCC.2.To study the protein expression of Cks1 and Cks2 in HCC.3.To study the relations between the expression of Cks1/Cks2 and the various clinicopathological features of HCC.4.To study the relations between Cksl/Cks2 and p27kip1 in HCC.Methods1.The mRNA expression of Cks1,Cks2 and p27kip1 were detected by real time PCR.2.The protein expression of Cks1,Cks2 and p27kip1 were detected by immuno -histochemistry.Results1.We successfully established relative quantization of real time PCR in detecting the mRNA expression of Cks1 and Cks2,which showed good specificity,perfect repeatability(CVs were all less than 3%),and high amplification efficiency (efficiency of amplification were 98.43%and 95.22%,respectively).2.Both gene and protein expression of Cks1 and Cks2 were elevated in HCC compared with the adjacent non-cancerous tissues(included chronic hepatitis and cirrhosis tissues) and normal liver tissues(P<0.05),but there showed no significant difference between the adjacent non-cancerous tissues and normal liver tissues(P>0.05).The mRNA expression of p27kip1 showed no significant difference between HCC and adjacent non-cancerous tissues(P>0.05),but its protein expression was lower in HCC than adjacent non-cancerous tissues(P< 0.05).3.Cks1 mRNA overexpression was associated with poor tumor differentiation, cancer embolus,tumor node and AFP(P<0.05),but it wasn't associated with age, sex,lymph node metastasis,tumor size and HBV(P>0.05);Its protein overexpression was associated with poor tumor differentiation,lymph node metastasis,tumor node and AFP(P<0.05),but it wasn't associated with age,sex, tumor size,cancer embolus and HBV(P>0.05).Cks2 mRNA overexpression was associated with sex,poor tumor differentiation,and AFP(P<0.05),but had nothing to do with age,lymph node metastasis,cancer embolus,tumor size,tumor node and HBV(P>0.05);Its protein over expression was associated with poor tumor differentiation and lymph node metastasis(P<0.05),but observed no relations with age,sex,cancer embolus,tumor node,tumor size,AFP and HBV(P >0.05).4.The expression of Cks1/Cks2 protein was negatively associated with p27kip1 protein(r1=-0.412,P=0.006,r2=-0.480,P=0.001).Conclusions1.Real time PCR is a goog tool to detect the mRNA expression of Cks1 and Cks2, which has showed good specificity,perfect repeatability,and high amplification efficiency.2.Cks1 and Cks2 were all elevated in HCC,and their overexpression were all associated with the degree of tumor malignancy.They may play an important role in the development of HCC.3.Cks1/Cks2 may promote the development of HCC throught p27kip1 protein.
