The Effects And Mechanisms Of 5-Fu On The Expression Of Transcriptional Factor OCT4 And SOX2 Of Gastric Cancer Cell

【Objective】To investigate the effects of 5一FU on proliferation inhibition in gastric cancer cell line SGC7901 and on transcription factor OCT4 and SOX2 related proteins and mRNA expressions. And to study the change of OCT4 and SOX2 signal pathway in gastric cancer cell line SGC7901 cells【Methods】SGC7901 cells were exposed to 5一FU at different dosages. Proliferation inhibition was detected by MTT assay. Culture gastric cancer cell line SGC7901,then detect the expression of OCT4 and SOX2 of transcription factor in these cell lines with immunohistochemistry. Furthermore, the expression of OCT4 and SOX2 of transcription factor gene was detected by RT-PCR.【Results】1.Expression of OCT4 and SOX2 transcription factor was analyzed by immunohistochemistry, gastric cancer cell line SGC7901 cells which were in an exponential stage of growth can express OCT4 and SOX2.The positive expression rate of OCT4 and SOX2 in SGC7901 cells stained intensity varyed from weak to strong.2.The results of immunohistochemistry and RT-PCR suggested that 5-Fu could enhance the expression of OCT4 and SOX2 of transcription factor in these cell lines.The results also suggested that it could enhance the expression of OCT4 and SOX2 of transcription factor in these cell lines bit by bit to booster dose and add time graduatly.【Conclusion】(1)Gastric cancer cell line SGC7901 cells could express core transcription factor OCT4 and SOX2. ( 2 ) 5一FU could enhance progressively the expression of core transcription factor OCT4 and SOX2 in these cell lines by boostering dose and adding time graduatly.
