Objects To investigate the effect of PHI on Burkitt lymphoma Daudi cell line in vitro, observing the state of epigenetics in Daudi cells after exposure to PHI, discussing the potential mechanism.Methods The viability of Daudi cells was observed by MTT method . Apoptotic rate was measured by flow cytometry. The variation of apoptosis associated protein Bcl-2, proCaspase-9, proCaspase-3, McL-1, XIAP was detected by Western Blot. The variation of acetylated H3 and H4, methylated H3K9 and H3K4, was detected by Western Blot.Results Compared with untreated cells, PHI induced decrease of cells proliferation and increase of cells apoptosis. It down regulated the expression of Daudi cells'apoptosis associated protein Bcl-2,McL-1,XIAP,released Cyt-C, activated proCaspase-9,proCaspase-3. PHI could induce the accumulation of histone acetylated H3,H4, increased histone methylation H3k4 and decreased methylated H3K9.Conclusions In vitro,PHI was able to up regulate the histone acetylation which associated with transcription stimulation. PHI specifically modulated histone methylation, inhibited methylation on histone H3 lysine 9, and raised methylation on H3 lysine 4, which were markers of activation of gene transcription. PHI could induced apoptosis through mitochondrion pathway and inhibited the cells growth in Burkitt lymphoma Daudi cell line. Briefly, PHI might be a potential new drug anti non-hodgkin lymphoma.
