| ObjectiveTo observe the change of the blood -pressure and the expression of theα-AR & ATR in the centre (brain)and periphery (kidney) after acute intracerebral hemorrhage(ICH) in SHR, study the relationship between blood-pressrue regulation and two types receptors, expression.MethodsA total of 30 Six-month-old male SHR was used , which were randomly divided into sham operation group and experimental groups according to Random number table, the experimental groups was further divided into bleeding 1d, 3d, 7d and 14d group ,each has 6 (according to random number from small to large order),the blood pressure was determined by tail-cuff, collagenase was injected into caudate-putamen nuceus to induce ICH model. Monitoring the change of blood pressure and ethology after operation. Decapitation to get the brain and the kidney at defined time, Immunohistochemistry and Reverse-Transcription polymerase chain reaction( RT-PCR) was performed to detect the expression of theα1A-AR/α2A-AR and AT1R/AT2R in the centre and periphery.Results1,The SHR appeared different degrees of behavioral change after ICH,the Bederson tests were nearly over 2 (2.33±0.701).2,Compared with before operation(177.8±8.69)and the sham operation group(184.1±3.76), the blood-pressure of SHR significantly statistical increased after ICH at the first day(195.4±8.39),(p=0.000,p=0.002),at the third day(185.3±9.22), it began to decrease and recover at 7d(177.7±5.62)~14d(176.7±6.06).3,Theα1A-AR mRNA and protein in the kidney were up-regulated after ICH at1d(0.91±0.013,0.944±0.142 %),especially at 3d (0.93±0.015,1.526±0.296%),compared with the sham operation group (0.89±0.018 , 0.779±0103%) , the differences were significant(p=0.008,p=0.010).4,Theα2A-AR mRNA and protein in the brain had significantly increased after ICH at 3d(0.93±0.020, 2.64±0.293 %) compared with the sham operation group (0.86±0.019,1.070±0.155 %)and 1d (0.87±0.029,1.629±0.488 %) ( p=0.001,0.020, p=0.000, 0.023).5,The expression of AT1R in the brain up-regulated after ICH, especially at 3d(p<0.05),and then recovered from 7 to 14d.6,AT2R in the brain and AT1R,AT2R in the kidney no difference were found after ICH.Conclusion1,the ICH animal model of collagenase injected into caudate- putamen nuceus in SHR was stable .2,The change of blood pressure in SHR after ICH was significant at the first 24h, it decreased at the third day and nearly return to before ICH at the 7d to 14d.3,the blood pressure increased after ICH in SHR maybe have some relationship with up-regulationα2A-AR in the brain andα1A-AR in the kidney. Also,it might relate with selectivity up-regulation AT1R in the brain. |
