Screening Liver Cirrhosis-Related Proteins In Serum By Liquid Chip-MALDI TOF Mass Spectrometry

Objective To screen liver cirrhosis-related proteins in serum by liquid chip-MALDI TOF Mass Spectrometry.Method The serum samples of 54 patients of chronic hepatitis B without severe liver cirrhosis(S≤1) and 54 patients of chronic hepatitis B with severe liver cirrhosis(S≥2)were randomly divided into two groups to build up the group of establishing model and the group of verifying model.The serum samples were detected by liquid chip-MALDI TOF Mass Spectrometry.The samples of establishing model were detected to screen the differential proteins and establish the diagnostic model.The differential proteins and the model were verified by the groups of verifying model.Result The serum protein spectrums of the group of establishingmodel including severe and light liver cirrhosis patients were compared and got 15 differential proteins.Among them,6 differential proteins were selected to establish the diagnostic model of liver cirrhosis,the m/z were 2069.67,2951.26,2209.86,1944.41,5248.41,5904.22 respectively.The sensitivity and specificity of the model was 100% respectively.The up-regulated proteins were 2951.26(m/z),5336.56(m/z),2555.98 (m/z),5248.41(m/z) 5904.22(m/z);The down-regulated protein was 1944.41 (m/z),2081.73(m/z),2209.86(m/z),4281.86(m/z),4475.06(m/z),3955.54 (m/z),3524.39(m/z),2069.67(m/z),3772.44(m/z),3813.28(m/z).Conclusion The differential proteins and diagnostic model were got by liquid chip-MALDI TOF Mass Spectrometry after the comparison of severe and light liver cirrhosis patients serums spectrum.The result provided new thought for diagnosis of liver cirrhosis.
