ObjectiveTo investigate the effects of herceptin on the proliferation and apoptosis of human endometrial adenocarcinoma Ishikawa cells in vitro and the probable mechanism invov led.MethodsMTT assay and flow cytometry were used to measure the effects of herceptin on cell proliferation and cell cycle;Western blot and ELISA were used to evaluate the ex pression of HER-2/neu protein and the expression of E2 in supernant at different time with different concentration of herceptin respectively.Results(1) herceptin inhibited cell proliferation of cultured Ishikawa cells in a dose dep endent manner(P<0.05);herceptin blocked the cells growth in G1-S period and incr eased the rates of cell apoptosis in a dose and time dependent manner(P<0.05);(3) herceptin decreased the expression of HER-2/neu protein in a dose and time depende nt manner(P<0.05).(4) herceptin decreased the expression of E2 in cell supernan t in a dose and time dependent manner.ConclusionsHerceptin is effective for inhibiting the growth of endometrial adenocarcinoma Ish ikawa cells.This anti-neoplastic effect may involve Blocking cell cycle in G1-S period, increasing cell apoptosis and decreasing the expression of HER-2/neu and E2 may inv olved in this anti-neoplastic effect of herceptin.
