Section 1 The effect of preeclampsia on physiology and pathology of fetal ratsObjective:To explore the effect of preeclampsia on physiology and pathology of fetal rats.Methods:Maternal rats are randomly divided into 3 groups:(1) high dose group:maternal rats are subcutaneously injected with 125mg/kg·d L-NAME in the 13th gestational day,and persisting in 7 days;(2) low dose group: maternal rats are subcutaneously injected with 75mg/kg·d L-NAME in the 13th gestational day,and persisting in 7 days;(3) control group:maternal rats are subcutaneously injected with 1 ml of 0.9%Nacl in the 13th gestational day,and persisting in 7 days.In the 21st gestational day, the maternal rats are operated in cesarean.And then maternal and fetal rats are separately phlebotomized.Then we mensurate the serum concentrations of ACTH,corticosterone and insulin of maternal and fetal rats.And we observe the pathology of adrenal gland and brain of fetal rats.Results:In the animal model of Preeclampsia,1.In the serum marks of maternal rats,the serum concentration of corticosterone of high dose group is significant lower than that of control group(P<0.05)and that of low dose group(P<0.01).the serum concentration of ACTH of high dose group is significant higher than that of control group and low dose group(P<0.01).2.In the serum marks of fetal rats,the serum concentration of corticosterone of high dose group is significant lower than that of control group and low dose group(P<0.05).The serum concentration of insulin of high dose group is significant lower than that of control group(P<0.05).3.In high dose group,the adrenal cortex of fetal rat is midrange hemorrhage and necrosis.The neuroglia is hyperplasia in brain cortex and medulla,and it is hyperplasia in hippocampus area,with hemorrhage.In low dose group,the adrenal cell is hyperplasia in adrenal cortex,with mild hemorrhage.The neuroglia is hyperplasia in brain cortex and medulla,and it is inflammatory infiltration in hippocampus area,with mild hyperplasia of neuroglia. Conclusion:Preeclampsia impaired the function of adrenal gland of maternal and fetal rats,the nerve cell and the function of pancreas of fetal rats.Section 2 The long-term effect of preeclampsia on adolescent offspring ratsObjective:To explore the effect of preeclampsia on cognitive development and neuroendocrine axis of adolescent offspring ratsMethods:Maternal rats are randomly divided into 3 groups like Section 1:high dose group,low dose group and control group.After the maternal rats is spontaneous delivery,the offspring rats are fed till they are 8 weeks old.And then some of the offspring rats are phlebotomized.Then we mensurate the serum concentrations of ACTH,corticosterone and insulin of the offspring rats.Some of the offspring rats were tested for visual - spatial learning and memory and spatial localization abilities in Morris water maze.Results:In the animal model of preeclampsia,(1) In the 1st day of place navigation test,the escape latent period of high dose group and low dose group is significant longer than control group(P<0.05).In the 4th day,the escape latent period of high dose group is significant longer than control group(P<0.05).In spatial exploration test,the percent ofⅣquadrant distance of low dose group is significant lower than control group(P<0.05). 2.The serum concentration of insulin of male offspring rats of high dose group is significant lower than that of control group(P<0.05).The serum concentration of corticosterone of male offspring rats of high dose group is significant lower than that of female offspring rats(P<0.05).The serum concentration of ACTH of male offspring rats of high dose group is significant lower than that of female offspring rats(P<0.05).The serum concentration of corticosterone of male offspring rats of low dose group is significant lower than that of female offspring rats(P<0.05).Conclusion:Preeclampsia results in the damage of intelligence of offspring rats.The study pattern has not significant difference.And memory and space direction ability of offspring rats is impaired.The fuction of adrenal gland of adolescent offspring rats recovers,but the fuction of pancreas of adolescent offspring rats does not recover.Section 3 The long-term effect of preeclampsia on adult offspring ratsObjective:To explore the effect of preeclampsia on blood pressure,blood glucose and cognitive development of adult offspring ratsMethods:Maternal rats are randomly divided into 2 groups like Section 2:high dose group and control group.After the maternal rats are spontaneous delivery,the offspring rats are separately fed.The offspring rats are detected blood pressure and blood glucose in five time point:the postnatal 100th day,120th day,150th day,180th day and Ist year. Then the 1-year-old offspring rats of these two groups were tested for visual - spatial learning and memory abilities in Morris water maze.Results:In animal model of preeclampsia,1.With the increasing of the age,the blood pressure of offspring rats of control group is gradually increasing.From the 120th day,the blood pressure of offspring rats of high dose group is significant lower than that of control group(P<0.05).2.The blood glucose of high dose group is significant higher than control group in the 120th,150th and 180th day(P<0.05).3.In place navigation test,the escape latent period of high dose group is significant longer than control group in the 2nd,3rd and 4th day(P<0.01).In spatial exploration test,the precent ofⅣquadrant distance has not significant difference between high dose group and control group(P<0.05).Conclusion:Preeclampsia results in changing the increasing pattern of blood pressure,increasing blood glucose and impairing cognitive development of offspring rats.Memory and space direction ability of offspring rats have not significant difference.
