The Study Of The Correlation Of AIB1 And P53 And Their Expression In Gastric Carcinoma

OBJECTIVE To study the clinical significances of the expression of AIB1(amplified in breast cancer 1)mRNA and protein in gastric carcinoma and to analyze the correlation of AIB1 protein and p53 protein.And to detect the mechanism of the growth and development of gastric carcinoma.METHODS Using semi-quantitative reverse transcription-polymerase chain reaction(RT-PCR),we examined the expression average quantity of AIB1mRNA and using immunohistochemistry,we examined the expression of AIB1 protein and p53 protein in 51 cases of gastric carcinoma and 47 cases of modest and severe chronic atrophic gastritis as well as 36 cases of chronic superficial gastritis(control group).The results were analysed for the expression of AIB1mRNA and protein and p53 protein,and the relationship with the clinical-pathological features.And to analyse the correlation of AIB1 and p53.RESULTS 1.The expression average quantity of AIB1mRNA(1) The comparison with gastric carcinoma group and modest and severe chronic atrophic gastritis group and control group.The expression average quantity of AIB1mRNA detected in gastric carcinoma group and modest and severe chronic atrophic gastritis group were significantly higher than that in control group,the differences had statistical significances(P＜0.01).And the expression average quantity of AIB1mRNA detected in gastric carcinoma group was significantly higher than that in modest and severe chronic atrophic gastritis group,the differences had statistical significances(P＜0.01).(2)The relationship between the expression average quantity of AIB1mRNA and the clinical-pathological features in gastric carcinoma group.The expression average quantity of AIB1mRNA was related to clinical TNM stage and lymph node metastasis(P＜0.05),but irrelevant to sex,age and pathology histological type(P＞0.05).2.The expression of AIB1 protein(1) The comparison with gastric carcinoma group and modest and severe chronic atrophic gastritis group and.control group.The positive expression rate of AIB1 protein detected in gastric carcinoma group and modest and severe chronic atrophic gastritis group were significantly higher than that in control group,the differences had statistical significances(P＜0.01 or P＜0.05).And the positive expression rate of AIB1 protein detected in gastric carcinoma group was significantly higher than that in modest and severe chronic atrophic gastritis group(P＜0.05).(2)The relationship between the expression of AIB1 protein and the clinical-pathological features in gastric carcinoma group.The expression of AIB1 protein was related to clinical TNM stage and lymph node metastasis(P＜0.05),but irrelevant to sex,age and pathology histological type(P＞0.05).3.The expression of p53 protein(1) The comparison with gastric carcinoma group and modest and severe chronic atrophic gastritis group and control group.The positive expression rate of p53 protein detected in gastric carcinoma group and modest and severe chronic atrophic gastritis group were significantly higher than that in control group,the differences have statistical significances(P＜0.01 or P＜0.05).And the positive expression rate of p53 protein detected in gastric carcinoma group was significantly higher than that in modest and severe chronic atrophic gastritis group(P＜0.05).(2) The relationship between the expression of p53 protein and the clinical-pathological features in gastric carcinoma group.The expression of p53 protein was related to clinical TNM stage and lymph node metastasis(P＜0.01 or P＜0.05),but irrelevant to sex,age and pathology histological type(P＞0.05).4.The correlation of the expression of AIB1 protein and p53 protein in gastric carcinoma group.The expression of AIB1 protein was related to p53 protein expression in gastric carcinoma group(P＜0.01).CONCLUSION 1.The expression of AIB1mRNA.and protein detected in control group was low level,and in modest and severe chronic atrophic gastritis group was middle level and in gastric carcinoma group was high level. Over-expressed AIB1 might be one of the factors promoting the development of gastric carcinoma.The expression of AIB1 was related to clinical TNM stage and lymph node metastasis,and it suggested AIB1 might be the important indicator of worse clinical prognosis of gastric carcinoma.2.The p53 protein detected in gastric carcinoma had higher positive expression rate,it participated with gastric carcinogenesis.The expression of p53 protein was related to clinical TNM stage and lymph node metastasis,it suggested p53 had some value about evaluating prognosis of gastric carcinoma.3.The expression of AIB1 protein showed positive correlations with p53 protein expression,both co-participated with the growth and the development of gastric carcinoma.
