Objective To observe the effect of transplantation of modifed PLGA complex substrate seeded with Schwann cells and neural stem cells to the injuried spinal cord in Wistar rats.Method PLGA was coated with Hydrophobin HFBI and then printed with fetal bovine serum to form channels.Then neural stem cells labeled with Brdu and schwann cells were seeded on modified PLGA to form a complex substrate.40 adult Wistar male rats weighing(200±30)g,were divided into 4 groups randomly:Control group(modified PLGA),SCs group(Schwann cells + modified PLGA),NSCs group(Neural stem cells + modified PLGA),and compound group(NSCs + SCs + modified PLGA).All these rats were hemi-sectioned on right spianl cord(T10).After that the complex substrate were twisted to form cylinder as a 3-dimension scaffold and then transplanted into injuried spinal cord to repair,respectively.After 8 weeks, observing functional recovery of injury spinal cord by BBB locomotion score, histopathology immunohistochemistry,immunocytochemistry,electrophysiological technique,HRP tracing and electron microscape.Result It was observed that after 8 weeks since the 3-dimension scaffold had been ransplanted the BBB scores in cells groups were higher than that in the control group(P<0.05).The recovery of motion function in compound groups was quicker than the other group(P<0.05).By HE tissue recovery and cells regeneration could be seen in the defected position in cells group but the control group could not be seen almost recovery or regeneration in the injuried site and the control group can be seeen scars formed.By sep,just after the operation,sep of all groups disappeared;After 8 weeks,all groups of sep recovered in different agrees.The compound group had the shortest potential latencies and the highest amplitude.By electrophysiological technique spinal cords of all groups in the detected position can conduct electric impluse.By HRP the compound group can be observed many stained nerve fibers and by Brdu could be observed a cell stained.By electron microscape,the compound group could be observed that neoformative and myelinated nerve fibers were found in the detected position and the control group could only be observed a few unmyelinated nerve fibers.Conclusion By seeded on the channels on the surface of the modified PLGA, NSCs and SCs can be conducted to adhere and grow oriented by the channels on the modified PLGA.Cells can almostly not adhere or even grow outside the channels. The modified PLGA seeded with cells were transplanted in injuried spinal cords.The channels can conduct axons growth oriented to connect the injuired areas of spinal cords.So the transplantation can improve the function recovery and tissue regeneration.The experiment presents important experimental evidence and new thought to treat spinal cord injury.
