AIM:To study the expression of Nanog gene in tumor-like stem cells of osteosarcoma and its signification.METHODS:Tumor sphere cells were isolated from osteosarcoma cell line U2-OS and MG-63 with serum-free culturing techniques,Stro+ cells were isolated using magnetic activated cell sorting system,RT-PCR was used to,detect the expression of Nanog gene in Stro-1+cells,Stro-1-cells and tumor sphere cells.RESULTS:The level of Nanog mRNA expression in stro-1+cells and tumor sphere cells 1.69±0.14,1.56±0.18 higher than the Stro-1-cells(P<0.05).Expression of Nanog mRNA in sphere cells were lower than the ealier time with the extension of culture(P<0.05 )CONCLUSIONS:Over expression of Nanog gene was observed in Osteosarcoma-like stem cells,this gene might have conduced to sustain the self-renewal and the undifferentiation of osteosarcoma-like stem cell.
