Exploring the active ingredient which has anti-radiation effect from natural products is a very important research topic.Polysaccharide is being a hot pot,as a kind of natural products with many physiological activities.Recently,most researches were about the fungal and plant polysaccharides,and it is found that many polysaccharides have anti-radiation effect,which is a kind of promising natural radioprotectors. Inspired by the physiological activities of these polysaccharides,we take into account of chondroitin sulfate which comes from animal,having wide range of resource,low cost,and easy to get.So we conduct the preliminary study to investigate protective effect of CS-A on damage caused by irradiation in mice.Effect of chondroitin sulfate-A on X-ray irradiated miceMETHODS:Male normal Kunming mice were fed with low,middle and high dose CS-A,and gave this treatment for 30 days.Then part of the mice was exposed to whole-body radiation and the total radiation dose was 5.0 Gy.24 hours later, periphery blood WBC,index of thymus and spleen,micronuclei frequency and DNA content in bone marrow of the mice in this part were measured.Single cell gel electrophoresis assay(SCGE) was used to monitor DNA damage in lymphocytes of periphery blood.And the other part of the mice were exposed to whole-body radiation and the total radiation dose was 8.0 Gy.After that,all the mice in this part were fed normally and the survival condition was observed.RESULTS.Low dose CS-A can resist the decrease of the periphery blood WBC.The three doses of CS-A all can decline the micronuclei frequency,decline the radiation damage on the DNA of lymphocytes in the periphery blood and prolong the mean survival time of the mice which received 8.0Gy X-ray.And the effect depended on the dose.However,the CS-A had no significant impact on the index of thymus and spleen.CONCLUSION:CS-A has certain protective effect on radiation-injury mice.This study provides the experimental basis for the development of radioprotectors and the new application of CS-A.Effect of chondroitin sulfate-A on heavy iron irradiation miceMETHODS:Male normal Kunming mice were fed with 0.2g·kg-1·d-1 CS-A and gave this treatment for 30 days.Then the mice were exposed to whole-body 12C6+ radiation and the total radiation dose was 2.0 Gy.After the radiation,all the mice were fed normally and stop giving CS-A.3d later,periphery blood WBC,index of thymus and spleen,DNA content in bone marrow,Superoxide dismutase(SOD) activity and Maleic dialdehyde(MDA) content in serum and liver and Glutathione(GSH) level in brain were determined.RESULTS:CS-A in this dose can resist the decrease of the index of thymus in irradiated mice.It can increase the DNA content in bone marrow,the activity of T-SOD in serum and live,and decrease the level of MDA in serum and liver.However, CS-A in this dose had no significant impact on the periphery blood WBC,the index of spleen and Glutathione(GSH) level in brain.CONCLUSION:To some extent,CS-A can inhibit the lipid peroxidation caused by radiation and protect DNA from the radiation damage.The results showed that CS-A has certain protective effect on heavy ion radiation-injury mice,and its mechanisms may be related to the two biological effects of CS-A,which are the anti-oxidation and the clearance of the free radicals.This experiment proved the certain protective effect of CS-A on the radiation mice further,and discussed the possible influence mechanism preliminary.It provided some experimental data for the biological effects of heavy ion irradiation on the normal mice at the same time.
