Background Human cytomegalovirus(HCMV)infection is universal and is one of the frequent conditional infection in man.Systemic lupus erthematosus(SLE) is a complex systemic autoimmune disease characterized by a diverse array of autoantibody production,complement activation,immune complex deposition,and tissue and oran damage. At present the cause and pathogenesis are unkown.Most people think genetic and environmental factors involve SLE. The rate of HCMV infection in SLE patients was higher than that in the healthy controls and HCMV infection is consider to induce SLE.SLE patients are easy to induce HCMV infection because of abnormal immune function and the use of glucocorticoid and immunosuppressant. The clinical manifestation of HCMV infection is multiformity,such as retinitis, interstitial pneumonia,hepatitis, thrombocytopenia.These manifestations are difficult to distinguish with the systemic damage caused by SLE,so they are incorrectly considered to be SLE disease flare.The use of large dose glucocorticoid and immunosuppressant leads to aggravate illness,even dead. So it is important to know the pathogenesis of SLE and evaluation illness through studying the relationship between patients with SLE and HCMV infection.Objectives To study the relationship between patients with systemic lupus erythematosus(SLE) and human cytomegalovirus(HCMV)infection.Methods HCMV isolation was assessed in the peripheral blood leucocyte (PBL) of 49 patients with SLE. Indirect immunofluorescence(IIF) was performed to investigate HCMV pp65 antigen and polymerase chain reaction(PCR) was performed to investigate the expression of HCMV UL54 DNA in cell cultures.Moreover,we detected HCMV DNA in PBL and urine of 65 SLE patients by fluorescence quantitative PCR.The clinical and laboratory parameters were also recorded. Results1,The rate of HCMV infection in SLE patients was higher than that in the healthy controls.2,Comparing the rate of HCMV infection, SLE patients in active phase were higher than those in inactive phase.3,The rate of HCMV infection in incipient SLE patients was higher than that in SLE patients who had cured, but they had no statistics difference.4,Total amount Urine protein in 24 hours,ESR were considerably higher in patients with positive HCMV infection than those with negative HCMV infection.5,Incidence rate of arthritis and positive rate of some autoantibody were considerably higher in patients with positive HCMV infection than those with negative HCMV infection.6,The rate of positive HCMV DNA in urine was higher than that in PBL by FQ-PCR.Conclusion1,The HCMV infection rate in SLE patients was higher than that in the healthy controls.2,HCMV infection might contribute to the disease flare in some SLE patients.3,HCMV infection was relative to Urine protein,ESR,incidence rate of arthritis and some autoantibody, it might induce SLE.4,When you detected HCMV DNA in SLE patients by FQ-PCR, urine specimen was better than PBL.
