Objective Discuss the relationship between the hyperlipidemia and the injury of vascular endothelial; detect the changes of the serum lipids, blood glucose, the liver histology and the cardiovascular ultrastructure in rat of obesity; observe the protective effect of different doses paeonol (Pae) on the vascular endothelial cells and the liver cells in rat of obesity.Methods 1)The 60 young SD rats were randomly divided into 6 groups, namely high-fat group (18), normal control group (18), high-dose paeonol intervention group (6), medium-dose paeonol intervention group (6), low-dose paeonol intervention group (6) and blank control group (6), in addition to the normal control group, other groups are set up obesity models in high-fat diet to rats; 2)Respectively, in the 4th week, 8th week and the 12th week to test the levels of serum glucose and blood lipids (TC, TG, VLDL) of rats in the high-fat group and the normal control group, in the 12th week to detect the levels of serum glucose and blood lipids (TC, TG, VLDL) of rats in the paeonol intervention group and blank control group; 3)Respectively, 2 rats in the high-fat group and the normal control group were killed in the 4th week, 8th week, 12th week, 2 rats in the paeonol intervention group and the blank control group were killed in the 12th week, respectively,to examine the vascular endothelial line of the myocardium, main coronary artery, thoracic aorta, etc. in the transmission electron microscope, and to examine the rats'liver sections after Sudanâ…¢staining in the light microscope.Results Compared with the normal control group, rats of the high-fat groups have significantly higher levels in the serum glucose, TC, TG, VLDL, etc. and body weight (P <0.05), the vascular endothelial line of the myocardial, coronary, aortic endothelial, etc. were obviously damaged, there is obvious fatty infiltration of liver; and compared with the high-fat group, rats of the paeonol intervention group have varying degrees of lower serum glucose, TC, TG, VLDL levels, etc. and body weight (P <0.05), and there is correlation between dose; rats'myocardium and coronary arteries, thoracic aorta endothelial, etc. to be a certain degree of protection in the electron microscopy; in light microscopy, fatty liver has been reversed to some extent.Conclusion High blood sugar and high blood lipids may be so obviously to damage the vascular endothelial; LDL modified by oxidation into ox-LDL, so that it produce vascular endothelial injury, HDL have protective function to vascular endothelial in effects. Paeonol inhibit low density lipoprotein (LDL) oxidative modification, inhibit the proliferation of vascular smooth muscle cell and play a protective role to endothelial cells, but also inhibit experimental atherosclerosis in the blood of animals, as well as the release of the inflammatory cytokines in some vascular tissue and have a dose-dependent manner, which may be related to decrease of contents of lipid, the extent of lipid peroxidation and the contentrations of proinflammatory cytokines. Paeonol tips the effects of inflammatory aspect of oxidized low-density lipoprotein-mediated vascular endothelial cells. Paeonol protect the liver and the hepatic fibrosis and its mechanism may also be able to significantly stimulate the plasma fibronectin levels and improve the liver microcirculation, reduce the pulse pressure of Portal vein, as well as improve plasma fibronectin (FN) levels, and reduce the liver cell degeneration and necrosis with interstitial inflammatory cell infiltration; Paeonol can improve the change of liver fibrosis in hepatitis, to promote the reabsorption of liver fibrous tissue.
