Objective 1 Try to investigate the relationship between CD4+CD25+ regulatory T cells express in peripheral blood and unexplained recurrent IVF-ET failure; 2 Try to investigate the relationship between Foxp3 express in peripheral blood and unexplained recurrent IVF-ET failure.Methods Unexplained recurrent IVF-ET failure women,normal non-pregnancy women and women succeeded in the first IVF-ET cycle were used for the experiment. The expression rate of CD4+CD25+ regulatory T cells was determined by double-label flow cytometric analysis: Immunohistochemistry was used for PBMC to know the expression of Foxp3; expression of Foxp3 mRNA was evaluated by RT-PCR.Results 1 The expression frequency of CD4+CD25+T cells and CD4+CD25highT cells were decreased significantly in unexplained recurrent IVF-ET failure women than in normal non-pregnancy women and women succeeded in the first IVF-ET cycle; 2 By immunohistochemistry, the expression of Foxp3 from peripheral blood was decreased significantly in unexplained recurrent IVF-ET failure women than in normal non-pregnancy women and women succeeded in the first IVF-ET cycle;3 expression of Foxp3 mRNA from peripheral blood of unexplained recurrent IVF-ET failure women was signifieantly lower compared with normal non-pregnancy women and women succeeded in the first IVF-ET cycle bv RT-PCR. Conclusion Downregulation of CD4+CD25high regulatory T cells and FOXP3 may be associated with unexplained recurrent IVF-ET failure. |
