Objective:To explore the role of CD4+CD25+CD127lo regulatory T cells and EL-10 in the pathogenesis of lupus nephritis,we detect the level of IL-10,CD4+CD25+CD127lo regulatory T cells(Tregs) in the peripheral blood of patients with lupus nephritis distinguished active stage and inactive stage.Other than,to respectively investigate the relationship between the level of Treg,IL-10 and Toxic Heat to Flourishing group,Deficiency of Yin group of LN patients.Methods:55 patients with lupus nephritis were differentiated according to various disease stage(active stage N=28,inactive stage N=27),and 21 cases of healthy volunteers were enrolled.Three-colour flow cytometry was used to quantitate proportions of Treg cells,and the level of IL-10 in the peripheral blood were detected by Cytometric Bead Array System.Results:1.Compared with the inactive stage of patients with lupus nephritis,the level of CD4+CD25+CD127loTregs from patients with active LN was lower(P<0.05),as well as compared the normal controls(P<0.01);compared with the normal controls,the level of CD4+CD25+CD 127lo Tregs from LN inactive stage patients was lower(P<0.05).2.Compared with the inactive stage of LN patients,the level of IL-10 was higher(P<0.01).3.The level of CD4+CD25+CD127lo T cell was correlated negatively with the level of IL-10 and SLEDAI (both P<0.01),and was not correlated with C3 and C4.4.SLEDAI was correlated positively with the level of IL-10(P<0.01).SLEDAI and the level of IL-10 were correlated negatively with C3 and C4 respectively(both P<0.01).5.Compared with the Deficiency of Yin group, the CD4+CD25+CD127loTregs of Toxic Heat to Flourishing group was lower(P<0.05),but the level of IL-10 was higher(P<0.01).Conclusions:1.The level changes of Treg and IL-10 in the peripheral blood of LN could be used as the index of lupus's reactiveness.2.There is negative correlation between Treg and IL-10 in the peripheral blood of LN patients.3.The level changes of Treg and IL-10 are involved in the pathogenesis of LN.4.The glucocorticoid hormone is helpful to elevate the level of Treg.5.In Zheng differentiation-classification of TCM,the levels of Treg and IL-10 are significant difference betweenin the toxic heat to flourishing group and Deficiency of Yin group,suggesting that the the levels of Treg and IL-10 of PBL of LN patients might be one of interal factors which formed different TCM syndrome types of LN.
