Objective: To investigate the expression of fucosyltransferase-8 (FUT8) in renal interstitial fibrosis(RIF) secondary to unilateral ureteral obstruction (UUO) in rats.Methods: In total, 90 healthy female wistar rats were randomly divided into 3 groups, 30 in each group: the normal control group(CON), sham operated group (SOR) and UUO group. The rats were randomly sacrificed on day 1,3, 7 ,14 and 21 after surgery, respectively. The histo- pathologic changes of renal interstitial tissues were observed with PAS and Masson staining. The expression of FUT8 was analyzed by immuno- fluorescence (IF), reverse transcription polymerase chain reaction (RT- PCR), and Western blot. Immunofluorescence costaining.was used to assess FUT8 and ALK5. The protein expressions of fibronectin (FN), type I collagen (colI) andα-smooth muscle actin (α-SMA) were analyzed by immunohistochemistry(IH) staining.Results: In the UUO group, the obstructed kidney showed typical features of progressive renal tubulointerstitial fibrosis.The level of FUT8 significantly increased at 7th,14th,21th day after the operation(P<0.05).The results of IF showed FUT8 and ALK5 coexpression in renal tubular and intestitial . As compared with CON group, FN ,colI andα-SMA, were significantly increased in the UUO group(P<0.05); and were positive correlation with the expression of FUT8 ( P<0.05) .Conclusion: Our data suggest that the expressions of FUT8 and ALK5 were time-space consistency increased in the development of renal interstitial fibrosis secondary to UUO in rats, and were closely correlated with renal interstitial fibrosis.
