LewisY antigen is a tumor-related carbohydrate antigen, residing in the codogenic production from erbB oncogene .These production are coded by epithelial growth factor receptor and immunoglobulin supergene family, regarding to tumorous malig- nant growth and regulation.LewisY antigen is overexpressed on the surface of human epithelial cancers. Monoclonal antibodies directed against LewisY antigen already succeed in manufacture, which can enhance antibody dependent cellular cyto-toxicity(ADCC).20(s)-ginsenoside is Chinese herbal drugs monomer microselected by the Red gindeng.During the past decade,many researchs confirmed that Rg3 have medicinal effects,such as anti-inflammatory,anti-aging activitiesand antitumorigenic.The mechanisam of its antitumorigenic effect is related to inhibiting angiogenesis,inducing tumor cell apoptosis and improving or- ganism immunity.Endometrial cancinoma is the most common gynaecological cancer in the Western world,and its incidence rate shows a raise trend in the world, obviously rising in our courtry.The Ratio of cervical cancinoma and endom- etrial cancinoma is upgrading from 8:1 to propter 1:1.The origin of endo- metrial cancinoma is endometrium glandular epithelium. It has been proved that LewisY antigen is 100% expressed in the tissue of endometrial can- cinoma and hyperplasia endometrium.It is not reported about targeted- LewisY antigen antibody therapy on endometrial cancer in home and abroad.We used the LewisY-specific anti- body combining 20(s)-ginsenoside(Rg3) to demonstrate the inhibitory effect on endometrial cancinoma cell growth in vitro.This study aimed to establish experimental evi- dence for the biotherapy of endometrial cancinoma.Objective:To investgate inhibitory effects of combining anti-LewisY antibody and 20(s)-ginsenoside on endometrial car- cinoma cellline A431 growth and reducing cell apoptosis in vitro.Methods:To divide the cell into the experimental group (LewisY- body, Rg3, LewisY-body+Rg3),and control group (A431) four groups.To compare the tumor control rate of each group by MTT ;to analyze each group cell cycle and apoptosis by FCM;to observe the cell morphology of cell apoptosis by hoechst33342 fluorescein stain and to detect the expression of VEGF in each group by immunohistochemistry and Western-blot.Results:1)The cell growth speed of each experimental group was slower than the control group,and the speed of the Le- wisY-body+Rg3 group was the most slowest,which depended on the cell growth time and drug-dose(P<0.05);2)in 48 hours ,the inhibitory effect of three experi- mental groups were different,the effect on LewisY-body+Rg3 group was the most obviously(P<0.05);3)the cell apoptosis rate of the control group, Le- wisY-body,Rg3 and LewisY-body+Rg3 were respectively 7.32%,13.09%, 12.57% and 24.91%.Comparing with LewisY- body+Rg3 and the other three groups respectively, the variability of cell cycle arrested in G1 phase was significan(tP<0.05).But there was not significant difference compared with each group in S and G2;4)the ex- pression of VEGF on the experimental groups were significantly down-regulated compared with the control one.Conclusion: Combining anti-LewisY antibody and Rg3 can inhibit the cell lines A431 growth as well as reduce tumour cell apoptosis.while they also down-regulated the expression of VEGF to refrain tumor angiogenesis.
