Objective: 1. Use modern inspecting measures to give an objective estimation about the treatment effect of"pneumonia mixture"against the klebsiella pneumoniae through animal experimentation. 2. provide evidence for the clinic application of"pneumonia mixture"combining with Chinese traditional medicine theory. 3. make a foundation for further utilizing of"pneumonia mixture".Pneumonia is kind of inflammation at the terminal respiratory tract, pulmonary alveoli and pulmonary interstitial caused by pathogenic microor- ganism, physico-chemical factors, damage of immunity, allergy and some medicines, classified into bacterial pneumonia, viral pneumonia, myco- plasma pneumonia, fungal pneumonia and pneumonia caused by other pathogens (rickettsia, chlamydia, toxoplasma, protozoon and parasite etc.), thereinto bacterial pneumonia is the most common and one of the most common infectious diseases as well. The pneumonia which is daily talked about refers to the one caused by bacterial infection. Bacterial pneumonia significantly threatened the health of children and the old before the application of antibiotics, which decreased the pneumonia death rate dramatically for a time. More powerful antibiotics and effective vaccines have been used in recent years nevertheless the total death rate caused by pnenumonia was still at the same level as before, even trending upward. Take the klebsiella pneumoniae as an example, its incidence has increased to 9%-37% in community acquired pneumonia and more than 70% in noso- comial acquired pneumonia. Though new antibiotics emerge in endlessly, due to the antibiotics resistance, the incidence rate of lung infection is still very high. The phenomenon of bacterial resistance, even multi-resistance appeared just after the application of a new antibiotics. For this reason, lots of medical staffs are now dedicated to the study on how to cure the above-mentioned infectious pneumonia with non-antibiotics, by which to defeat this difficult problem at last.We were delighted to find that the"pneumonia mixture"which was used by Chinese traditional professors for more than a decade has characters of fast effect, short course of disease and non-relapse against acute or chronic pneumonia, or the middle-aged and aged pneumonia or children pneumonia, which proved it was more effective than antibiotics. To probe into its fuc- tional mechanism, the klebsiella pneumoniae with high drug resistance character was chosen as the object in this research, in which we made a tentative study on pneumonia mixture's anti-inflammatory and anti-bac- terial effects by the method of infecting mice's lung with the klebsiella pneumoniae.Methods: The experimentation is divided into three parts, 1. the estab- lishment of mice model with bacterial pneumonia; 2. comparison of patho- logical sections of lung tissues after gavage medication; 3. bacterial culture of gavage medication after modeling.Results: The experimentation result showed that each Chinese tradi- tional medicine groups had the anti-inflammatory and anti-bacterial effects. In the observation of treatment effect against bacterial pneumonia, we found that compared with model groups, the pneumonia mixture high-dosage group and group were more effective to protect mice, prolong their lives and decrease their death rate (survival rate of mice P<0.05, survival days P<0.01) .When observing the pathological section of lung tissues with electron microscope, we found the phenomenon that ciliated columnar epithelium on the inner wall of bronchiole were clearly showed with no hyperplasia on the wall, no effusion from pulmonary alveoli and no inflammatory soakage in the normal group, however, in the model group, we found the phenomenon of neutrophils effused from the surrounding of bronchium and bronchiole and pulmonary alveoli and inflammatory soakage which could be judged as bronchopneumonia in different degrees, which indicated that the establi- shment of mice model with bacterial pneumonia became successful. When comparing the treatment group with the model group, we also found that the inflammation of pneumonia has been eased, the cell hyperplasia and inflammatory effusion abated, which indicated that each Chinese tradional medicine group had anti-inflammatory and anti-effusion effects. In lung bacterial pure culture of model group and other three treatment groups, the klebsiella pneumoniae were positive, while the control group were negative. In terms of the anti-bacterial effect of pneumonia mixture outside mice's body, we found that it's MIC50 and MIC90 were 0.125 g/ml and 1.000 g/ml respectively, and the scope of MIC was from 0.030 g/ml to 1.000 g/ml.Conlusion:"pneumonia mixture"can ease the damage of lung tissues, improve the condition and has definite effect to cure the mice with the klebsiella pneumoniae.
