Objectives NKG2D are primarily activated receptors expressed in all the NK cell surface which mediated NK cell recognitize and lysis tumor cells. NKG2D ligands expressed on a variety of tumor cells, and in retinoblastoma expression has not been reported. In this study, the expression of NKG2D ligands are detected on RB cells, and the expression changes of NKG2D ligands when ultrasound-mediated P53 gene transfected into RB cells in the RB to provide new ideas for immune research.Methods1.In this experiment,the expression of NKG2D ligands were measured by reverse transcription-polymerase chain reaction(RT-PCR).2. The cytotoxicity of natural killer(NK) cells were detected by MTT assay .3.Ultrasound-mediated P53 gene transfected into RB cells under the conditions of 0.5W/cm2 sound intensity, irradiation time 30s, continuous wave of ultrasonic .4.The expression of NKG2D ligands were measured by reverse transcription-polymerase chain reaction when transfected P53 gene into RB cells.5. RB cell apoptosis are detected by Flow cytometry .Resluts1. It was found that MICA,MICB,ULBP2,ULBP3 was expressed on RB cell,ULBP1 were not detectable on RB;K562 expressed all the NKG2D ligands.NKG2D ligands were highly expressed on K562 cell than RB cell(P<0.05). NK cells displayed highly in vitro cytotoxicity against K562 and RB cells (p<0.05).2. MICA,MICB,ULBP2 were expressed on RB cells when transfected with P53 gene, transfected groop expressed lower than blank groop,and the RB cell apoptosis in the two groop are the opposite (p<0.05).Conclusion1. NKG2D ligands also expressed on the RB cells.2. Expression of NKG2D ligands is correlated with the cytotoxicity of NK cells,ULBP1and ULBP3 may play an important role in cytotoxicity of NK cell.3. Transfected with P53 gene made tumor cell apoptosis and cut down the expression of NKG2D ligands in tumor cells.
