Negative Pressure Wound Therapy Upregulates Hypoxia Inducible Factor-1α And VEGF Expressions During Chronic Wound Healing

Objective The possible mechanism of NPWT to promote the growth of Chronic wound blood vessels has been studied by observing the impact of negative pressure wound therapy(NPWT) on the wound and its circum. Methods 12 patients with chronic wounds were selected from our hospital, then they were randomly divided into experimental and control group. The experimental group were treated with NPWT whereas the control group were treated with the traditional method , namely conventional dressing with NS dressing. Full-thickness skin on the verge of the wound were collceted respectively in the 1st before the treatment and in the 1st, 2nd and 7th days after the treatment. The amount of the hypoxia inducible factor-1α-mRNA(HIF-1α-mRNA) were detected by Reverse transcription polymerase chain reaction(RT-PCR).The expression of HIF-1-αand VEGF were detected by Immunohistochemistry(IHC) methed. Results The expression of HIF-1-α-mRNA of the experimental group grows more than that of the control group, whose difference were statistically significant (P <0.05).The expression of HIF-1αof the experimental group grows more than that of the control group, whose difference were statistically significant (P <0.05). The expression of VEGF of the experimental group grows more than that of the control group, whose difference were statistically significant (P <0.05). Meanwhile, the expression of VEGF and the expression of HIF-1αwas positively correlated(r=0.854,p<0.01,n=48). Conclusion NPWT can induce a high expression of HIF-1α,then increase the expression of VEGF to promote wound blood vessel growth and consequently to promote wound healing.
