Objective: To investigate the inhibitive effect of danshensu on JNK signaling in activated rat Hepatic stellate cells(HSCs),to explore the anti-hepatic fibrosis mechanisms of danshensu.Method: HSCs were isolated from rats . MTT colorimetric assay was used to detect proliferation of HSCs treated with different concentration of danshensu. The expression of JNK and P-JNK was observed by westernblotting assay when HSCs stimulated by IL-1βwas incubated with danshensu.Synthesis and secretion of collagenⅠwere detected by the quantitative immunocytochemical assay and ELISA method espectively.Results: Compared with the 0mg/ml group,the proliferation of HSCs in other concentration groups was significantly inhibited by Danshensu in a dose dependent manner.Danshensu in the concentration of (0.0625~0.25mmol/L) could prevent the proliferation of HSCs induced by IL-1βin a dose-dependenting manner(P<0.05). The concentration of 0.25 mmol/L Danshensu could downregulate the synthesization and secretion of TypeⅠc ollagen at 24h in HSCs,with or without stimulation of IL-1β(P<0.01).P-JNK expression of HSCs stimulated by IL-1βwas reinforced , Danshensu could downregulate P-JNK expression of HSCs(P<0.05),but had no obvious effect on the expression of JNK.Conclusions: Danshensu could inhibit the activation and proliferation of HSCs,and reduce the synthesization and secretion of TypeⅠcollagen .The mechanism may be concerned with danshensu inhibiting the JNK Signal transduction.
