| Ganoderma lucidum possess various active ingredients and comprehensive biological functions. It has considerable therapeutical effect to human nervous system, breathing system, incretion and metabolizing system. It has also displayed different activities including improving immunity, protective hepatic cells and deintoxication, hypoglycemia, antitumor and anti-senium. Accordingly, it has extensive application in the areas of medicine and clinic, health care foods, cosmetic and so on.Electrochemical sensor and DNA electrochemical biosensor have been used many fields such as food industry, disease diagnosis, pharmaceutical analysis and environmental monitoring due to the high sensitivity, rapid response, easy handling, no pollution and low cost. However, the evaluation of antioxidant activity of Ganoderma lucidum by electrochemical technology has not been well documented. In order to study and explore the resource of Taishan Mountain Ganoderma lucidum, the antioxidant activity of Taishan Mountain Ganoderma lucidum was studied by electrochemical technology. The result laid the foundation of correlative theories and application for the product research of Taishan Mountain Ganoderma lucidum. The main results and conclusions are as follows:(1) The modified electrode of iron cyanide gadolinium was prepared by electrochemical deposition, and the electrocatalytic oxidation of guanine on this modified electrode was studied. The scavenging effect of Vc for hydroxyl radicals was evaluated based on competitive reactive between guanine oxidation with hydroxyl radicals generated in Fenton system and guanine oxidation on the electrode directly. The results demonstrated that the scavenging effect of Vc for hydroxyl radicals was good within the concentration of Vc from 0 mmol/L to 2.5 mmol/L range, and this mothod is suitable for assessment of antioxidant capacity of antioxidant.(2) The modified electrode of iron cyanide terbium was prepared by electrochemical deposition.The antioxidation capacity of Taishan Mountain Ganoderma lucidum extracts by different solvents was evaluated based on the modified electrode. The results showed that the scavenging capability to ?OH was best, no matter what water extracts, 95% ethanol extracts and chloroform extracts of Ganoderma lucidum. It is showed by compared, the antioxidant capability of 95% ethanol Ganoderma lucidum extracts was best, then was that of water extracts and last was chloroform extracts. The IC50 values of 95% ethanol extracts, water extracts and chloroform extracts were 0.396 mg/mL, 0.443 mg/mL and 0.507 mg/mL, respectively.(3) A novel DNA electrochemical biosensor was prepared using dsDNA immobilized on PAMAM/CoHCF modified electrode, which was assembled G4 PAMAM on the CoHCF surface using the electrostatic interaction. The DNA electrochemical biosensor was characterized by electrochemical impedance spectroscopy. It is showed that the amount of the dsDNA on the modified electrode was increased, and the sensitivity of the biosensor was also improved. The antioxidant property of Ganoderma lucidum extracts of different source was evaluated using thie DNA electrochemical biosensor. The results showed that the antioxidant property of Taishan Mountain Ganoderma lucidum were very good, and there was no different with that of other source. |
