Objective: To establish SD rat model of asthmatic airway remodeling and investigate the role of oxidative stress in airway remodeling and airway inflammation. To analyze the impact of the antioxidant tea polyphenols, Chinese medicine Cordyceps sinensis(Jinshuibao) on airway inflammation, airway remodeling. To research for new treatment in airway remodeling of asthma.Methods: 1,Using ovalbumin (OVA) plus immune adjuvant (aluminum hydroxide gel) to establish rat model of chronic asthmatic airway remodeling by lenthing OVA challenge time. Cell counts and classification of BALF, score to inflammatory index around bronchial and the goblet cell mucus index, smooth muscle area of bronchial wall(WAm), collagen deposition area of bronchial wall (Wcol), integral optical density values (IOD values) of TGF-?1 of lung tissue were observed in 4 weeks, 8 weeks, 12 weeks and 16 weeks.2,Based on rat model of airway remodeling in asthma, to explore the impact of tea polyphenols, chinese medicine soft capsule Cordyceps sinensis on oxidative stress, airway inflammation and airway remodeling. TGF-?1, malondialdehyde(MDA), and superoxide dismutase (SOD) of lung tissue were observed.Results:1,Airway remodeling occurred significantly in early stage in asthmatic rat and the more these changes over time, At same time, the numbers of Eos gradually decreased, PMN gradually increasing.2,After the tea polyphenols or Cordyceps intervention, the oxidative stress level, airway inflammation, and airway remodeling significantly improved. Early intervention is superior to late intervention and the effect of high-dose of cordyceps is better than that of low-dose. The effects of budesonide alone were inferior to the tea polyphenols or Cordyceps in late intevention.Conclution:1,The method used 7 days interval allergens sensitized and extend challenge time by ovalbumin aerosol can be successfully established a good SD rat model of airway remodeling in asthma. PMN gradually increased in the late stage. 2,Oxidative stress may play a role in asthmatic airway remodeling. 3,Tea polyphenols, cordyceps may delay the formation and development of airway remodeling by controlling the level of oxidative stress and reducing airway inflammation (especially PMN), suppressing the expression of TGF-?1. Early drugs intervention is superior to late intervention. The effects of high-dose cordyceps is better than that of low-dose . The effects of budesonide alone were inferior to in late intevention.
