Objective:To establish ovarian cancer animal moldel, give the different small dose triptolide to them. After that, observe the tumor growth and immunologic function change of ovarian cancer rat after treatment. Investigating the effect the small dose triptolide on tumor growth and immune function of ovarian cancer rat in order to provide the experiment based on for Trip treating the ovarian cancer patient.Methods:To culture in vitro inbred strain Fischer344 rat poorly differentiated epithelium tissue ovarian cancer cell line NUTU-19. It take 8 in random for the normal groups in 48 F344 rats. Other 40 rat, it was transplanted 2×106 cell/merely at the rat right armpit hypo, establishes the ovarian cancer hypodermic transplanting tumor model rat. When the tumor of rat right armpit growers about 0.4×0.4 cm2 around, it was divided into 5 groups in random; Blank bearing cancer group: Each intraperitoneal injection rats with 2ml normal saline; DDP group: 3mg/kg.d of cisplatin by 2ml normal saline intraperitoneal injection rats; Trip A group: 0.1mg/kg.d of Trip by 2ml normal saline intraperitoneal injection rats; Trip B group: 0.05mg/kg.d of Trip by 2ml normal saline intraperitoneal injection rats; Trip C group: 0.025mg/kg.d of Trip by 2ml normal saline intraperitoneal injection rats; Every one day one time and medication for 5 secondary. To kill each group rats after 24 hours when the last medication, and use weighing method and calculating inhibitory tumor rate, and apply MTT assay and detect spleen lymphocyte transformation rate of rats, apply Elisa assay and detect TNF-a and IL-2 of rats blood-serum.Result:1. The effect of the Trip to rats tumor growth: Trip A group,Trip B group and Trip C group to ovaril tumor control rate respectively is 39.13%, 31.10%, 22.76%. the tumor control rate of DDP is 42.13%. Trip A group and DDP group were significantly differences than blank bearing tumor group(p<0.05). Trip B group and Trip C group were not significantly differences than blank bearing tumor group (p<0.05). Trip A group were not significantly differences than DDP group。2. Effect of the trip to the rat's immunologic function influence:(1) The spleen lymphocyte transformation rate of Trip A group,Trip B group,Trip C group and DDP group respectively is 1.165±0.04,1.181±0.06,1.208±0.07,1.157±0.04,that is lower than blank bearing tumor group(1.254±0.07), the difference has statistics significance(P<0.05). Each Trip group is not statistically significant than DDP group(P>0.05).(2) The IL-2 of the rats blood serum of Trip A group,Trip B group,Trip C group and DDP group respectively is 70.38±6.89ρg/ml,85.73±8.38ρg/ml,92.06±10.01ρg/ml,74.49±11.96ρg/ml,that is lower than blank bearing tumor group (108.94±18.90ρg/ml), the difference has statistics significance(P<0.05). Trip A group,B group is not statistically significant than DDP group(P>0.05). Trip C group is statistically significant than DDP group(P<0.05).(3) The TNF-a of the rats blood serum of Trip A group,Trip B group,Trip C group and DDP group respectively is 147.29±29.33ρg/ml,187.09±30.67ρg/ml,198.64±28.37ρg/ml,118.98±25.48ρg/ml,that is lower than blank bearing tumor group (228.84±32.56ρg/ml), the difference has statistics significance(P<0.05). Trip A group is not statistically significant than DDP group (P>0.05). Trip B group,C group is not statistically significant than DDP group (P>0.05).Conclusion:(1) The Trip has the inhibitory action to the ovarian cancer rat tumor's growth. The effect of the 0.1mg/kg of Trip restrain tumor is same the DDP. Thus, the Trip was be use the DDP-fast overian cancer's treatment.(2) The small dose Trip has the inhibitory action to the ovarian tumor rat spleen cell transformation function and cut down the IL-2 and TNF-a of the rat blood serum. The effect of the Trip immunction is not significant difference with the DDP, and it possibly becomes that the Trip treat the DDP-fast overian cancer .(3) The research take the ovarian cancer rat as the animal model, discovered that the Trip has obvious inhibitory action to the ovarian cancer, and the effect of the Trip immunction is not significant difference with the DDP, and also offered the animal experimentation foundation for the Trip treats the ovarian cancer patient.
