Objective: To investigatet the expression and distribution of monocyte chemoattractant protein-1(MCP-1) and the macrophage infiltration in oral submucous fibrosis(OSF), and further discuss their possible role in pathology of OSF and the relationship between the expression of MCP-land the macrophage infiltration in OSF.Methods: The specific SP immunohistochemistry was used for detecting the express and distribution of MCP-1 and the macrophage marker-CD68 in tissues of 15 cases normal oral mucosa and 45 cases OSF. The positive reaction and staining intensity were observed and counted blindly.Results: (1)A slight positive expression of MCP-1 protein was detected in normal oral mucosa, and a significant increasing of expression could be found in every stage OSF(P<0.01). Compared with intermediate and advanced stages, the expression of MCP-1 of early stage OSF was highest (P<0. 01), no significant difference was found in the expression between the intermediate and advanced stage 0SF(P>0..05).(2) A sprinkle macrophage was seen in normal oral mucosa, and a significant increasing of expression could be found in early, intermediate, advanced stage OSF. A significant difference was between every stage OSF and normol control gruop(P<0. 01). Compared with intermediate, advanced stages, the quantity of macrophage of early stage OSF was highest (P<0.01), the quantity of distribution between the intermediate and advanced stage OSF showed no statistical difference (P>0. 05).(3) The quantity of infiltrited macrophage in OSF were accompany with the expression of MCP-1. With the intensisity grade of MCP-1, the quantity of macrophage was respectively (48.07±5.325), (54.00±5.638), (66.70±4.001), which was increased gradually (P<0. 05).Conclusions: The upgrade expression of MCP-1 and macrophage both have been found in OSF, and the early stage was the highest. MCP-1 was the specific chemotaxin of monocyte/macrophage which may influence macrophage infiltration in OSF.
