Objective: To study the bone marrow-derived mesenchymal stem cell transplantation combined Neurotrophin -3 of the rat corticospinal tract transection and neuronal apoptosis after spinal cord injury and repair significance.Methods: Using density gradient centrifugation and filter combined adherent rat bone marrow-derived mesenchymal stem cells, selection of two hundred adult health Sprague-Dawley(SD) rats, weight 250-300g, male and female open, equivalent to the level of T8 in the lamina marked with micro-scissors cut the spinal cord of the dorsal 2 / 3, the establishment of rat corticospinal tract transection injury model. Surgery were randomly divided into control group (n = 50), NT-3 group (n = 50), BMSCs transplantation group (n = 50), BMSCs transplantation combined with NT-3 group (n = 50). 5 each in check, before surgery and after surgery 1d, 1w, 2w, 3w and 4w, the use of BBB (Basso, Beattle, Bresnahan) score assessment of motor function, and then remove the spinal cord to detect the molecular weight of 200,000 nerve microfilament proteins (neurofilaments, NF) immunohistochemical changes in the activity.The rest of the control group and BMSCs transplantation group, BMSCs transplantation combined with NT-3 group, NT-3 group of animals after injury to the 2h, 4h, 6h, 8h, 12h, 1d, 3d, 7d and 14d removed spinal cord injury, respectively, RT-PCR (reverse transcription-polymerase chain reaction) detection of bax and bcl-xl gene expression changes, at each time point five rats. The data used are the number of inter-group comparison t-test and analysis of variance group for statistical analysis.Results: dual-hindlimb after spinal cord injury paralysis, BBB score, 2w, 3w and 4w point of NT-3 group, BMSCs transplantation group, BMSCs transplantation combined with NT-3 group was significantly higher than the injury transplant control group, statistically significant differences (P <0.05); BMSCs transplantation combined with NT-3 group was higher than NT-3 group, BMSCs transplantation group, there are significant difference (P <0.05); (2) injury bax and bcl-xl gene mRNA transcription levels, However, more pronounced increase in bax gene; 3, NT-3 group, BMSCs transplantation group compared with the surgical control group, reduction in the level of bax mRNA and bcl-xl mRNA levels (P <0.05); BMSCs transplantation combined with NT-3 group and NT-3 group, BMSCs transplantation group than the reduction in the level of bax mRNA and bcl-xl mRNA levels increased significantly (P <0.05). 4, NF immunohistochemistry results showed that, BMSCs transplantation combined with NT-3 group was significantly less than the average gray the other three groups, statistically significant differences (P <0.05).Conclusion: (1) after spinal cord injury increase the expression of NF and NT-3, BMSCs can promote NF expression; (2) NT -3, BMSCs may affect the apoptotic gene bax and bcl-xl expression changes in spinal cord injury and repair protective role. (3) BMSCs transplantation combined with NT-3 in spinal cord injury can promote the survival and regeneration of nerve and synergy with each other over a single application of BMSCs or NT-3 to better promote the repair of spinal cord injury and functional recovery of rat hindlimb.
