Background and Objects:Alzheier's disease,a centra neurodegenerative disease which is characterized by evident learning,memory and cognitive function impairment.The pathology characteristics are the deposition outside the neuron cell of senile plaque(SP) which is mainly composed ofβ-amoyloid protein,inside the neuron cell of neurofibrillary tangles and the absence of neurons and axons in hippocampus,cerebral area.Although it's etiopathogenisis and mechanism is still unknown,neuro-inflammation from deposition ofβ-amoyloid has been accepted as a pathological core for Alzheier's disease.There are many Chinese crude drugs which are good for the therapy of AD.This text approach Nao-ling Decoction take part in the change of AD model rats'learning,memory and pathological,and the effect on OX-42 and IL-6,which lays the foundation for further elucidating its mechanism and treatments for AD.Methods:There were 40 pure line SD rats,then they were divided in random into 5 groups:a normal group,a sham operation group,a model group,a wester medicine group,Nao-ling Decoction group(that was traditional chinese medicine group,TCM group).each group is 8 rats.After feed a week,the AD model rats were established by injectionβ-amoyloid 1-42(Aβ1-42) into hippocampus,then feed three weeks.After they were cured for four weeks,their ability of learning and memory were detected by Y-maze and Morris water maze test.The brain tissue of rats were removed to be made frozen sections,the deposition ofβ-amoyloid protein was shown by Congo red stain,the changes of cell morphology were detected by HE stain and Nissl stain,the expression of OX-42 and IL-6 in hippocampus was measured with immunohistochemical stain.Results:1.Y-maze test:The times needed to learn how to evade electric stimulation in the TCM group rats and the wester medicine group rats were significantly less than those in the model group(P<0.05).Morris water maze test:①Scoured experiment on delitescence platform:the escaped and latent period of the model group was mostly delayed than other groups.compared with the TCM group and the wester medicine group,there were significant difference(P<0.05).②exploratory experiment:the average pass through times in normal group and sham operation group were more than that of other ones,and that of the model group was lest,there were significant difference(P<0.05).2.Three stains:①Congo red stain:Although there was deposition ofβ-amoyloid protein both in model group,wester medicine group and TCM group hippocampus sector,there was obviously less in TCM group and wester medicine group.②HE and Nissl stain:In normal group and sham operation group rats,the number of neurons in hippocampus CA3 sector were not loss and spaced neatly;In model group rats,the number of neurons in hippocampus CA3 sector were less and the array of neurons was more irregular than those in TCM group and wester medicine group.3.compared with the masculine cells about OX-42 and IL-6 in each group,the model group was the most,the TCM group and wester medicine group were more than the normal group and sham operation group(P<0.05),they had stastisical significance.The compassion of the gray scale value about IL-6 in hippocampus CA3 sector of each group,the model group were lower than the normal group and sham operation group,the gray scale value of TCM group and wester medicine group were higher than the model group(P<0.05),they had stastisical significance.Conclusions:1.The presentation of results of the study that the model was established by Aβ1—42 injected into hippocamal was succsesful,because the model rats had the changes of praxiology and pathology.2.The result of Y-maze test and Morris water maze test:the learning and memory of the model rats were decreased obviously,Naoling Decoction can improve the learning and memory of the model rats.3.Naoling Decoction can dcrease neuronal loss and better pathomorphological changes in hippocampal CA3 of AD rats,thusly it can prevent neurons in hippocampus CA3 sector of AD rats from impairment.4.Naoling Decoction can weaken the expression of OX-42 and IL-6 in hippocampal CA3 of AD rats,those partly may be therapeutic mechanism on Alzheimer disease of Naoling Decoction.
