Objective:1:To investigate the effect of proliferation inhibit and radiosensitivity of Tripterine on Nasopharyngeal Carcinoma(CNE-1)cell in-vi tro and its mechanism。2:To provide the experimental evidence for Tripterines' Anti-Carcinoma effect and finding new RadiosensitivityMethods:The cell-line were divided into groups depend on requestment,The inhibition ratio of Tripterine on Nasopharyngeal Carcinoma(CNE-1)cell was determined with MTT:the morphological changes of cells were observed by both light microscope and transmission electron microscope;,The radiosensitity(plot survival curv) was evaluated with Colony forming assay and Muiti-hit single-target model;Cell cycle distribution after Tripterine treatment was analyzed by using flowcytometry(FCM).Immunochemistry was used to determine expression of Fas-proteinResults;In certain range of concertration.Tripterine could cause inhibition proliferation of CNE-1 cells significantly in a dose and time dependent manner,The Medial Lethal Dose(IC50) was 32μg/mL;In result of light microscope and TEM,microvillus disappered,chromatin condensed to high electron density,nucleus broken to pieces and large deciduous apoptosis bodies appeared after being treated with Tripterine;Cell survival expeirment and plot survival curv showed the increase of SER and decrease of Do and Dq when CNE-1 cells were irradiated with Tripterine;Tripterine definitely enhanced radiosensitivity of CNE-1 cells.The outcome of FCM indicated that TriPterine afact obviously cell-cycle distribute and induced efficiently G1 and G2/M phase arrestin this cell-line following 48 hours of exposure which was in dose dependent manner.Meanwhile Immunochemistry show that the expression of Fas-protein increased after Tripterine treatment in dose dependent.Conclusion:1.Tripterine could inhibit the growth of Nasopharyngeal Carcinoma CNE-1 celllines significantly and increase the radiosensitivity of it。2.The mechanism of radiosensitivity may be associated with blocking cell cycle and inducing apoptosis increasing the expression of Fas-protein.
