BACKROUNDSMigraine is a kind of common clinical non-organic headache,but its mechanism remains not clear enough.The theory of brainstem-trigeminal nerve-vasculitis is becoming the focus of research on migraine these years,which posits that its main pathophysiology is meningeal vasodilation and inflammatory reaction with abnormality of many inflammatory cytokines like interleukin-1β(IL-1β),interleukin-2 interleukin-6,tumor necrosis factor(TNF) and monocyte chemoattractant protein-1(MCP-1).Some researches suggest that interleukin-17 can activate and up-regulate inducing-nitricoxide synthase(iNOS) followed by production of nitricoxide,interleukin-1,tumor necrosis factor and monocyte chemoattractant protein-1,which just closely related to migraine.Thus, we assume that IL-7 may take part in migraine's mechanism,and there is no such report so far in the world.It has been proved that nuclear factor-kappa B(NF-κB),a factor that plays an important role in inflammation,has something to do with migraine in experimental migraine model induced by nitroglycerin. However,there is no report about the effect of NF-κB on migraineurs and relationship between it and TNF-α,IL-17. ObjectivesTo observe changes of TNF-αand IL-17 in culture supernatant which is from monocyte cell line being cultured in migraineur's serum in vitro,and the effect of NF-KB inhibitor.To study the possible mechanism of NF-KB and relationship with TNF-αand IL-17.MethodsSelect 40 migraineurs which fit international diagnostic criteria of migraine and 40 normal people as control group.The main experimental objectives were normal and migraineur's serum and monocyte cell line of human(THP-1).THP-1 cells were cultured in 10%fetal calf serum plus 1640 medium which contains 100U/ml penicillin and 100ug/ml streptomycin.THP-1 cells were divided into 3 groups:normal control group,migraineur's serum group and migraineur's serum plus BAY11-7082 group.After 24hrs' being cultured in migraineur's serum,the TNF-αand IL-17 content were measured by ELISA,and do Pearson liner correlation analysis.Results1.After being cultured for 24hrs in migraineur's serum,TNF-αand IL-17 in culture supernatant of THP-1 cells are more than control group, and there is statistical significance(p<0.05).2.Culture supernatant of THP-1 cells express lower level of TNF-αin migraineur's serum group than in migraineur's serum plus BAY11-7082 group,and there is statistical significance(p<0.05).3.Culture supernatant of THP-1 cells express lower level of IL-17 in migraineur's serum plus BAY11-7082 group than in migraineur's serum group,and there is no statistical significance(p>0.05).4.Correlation analysis reveals:TNF-αis significantly correlated with IL-17 in migraineur's serum group(r=0.906,P<0.01).TNF-αis significantly correlated with IL-17 in migraineur's serum plus BAY11-7082 group(r=0.871,P<0.01).TNF-αis significantly correlated with IL-17 in nomal control group(r=0.885,P<0.01)ConclusionsAt the stage of attack,there possibly is increase of inflammatory factors TNF-αand IL-17 induced by NF-κB in migraineurs.
