Objective: To investigate the relationship of CD34+,Interleukin-5,Interleukin- 5Rαin the bone marrow and lung tissue of asthmatic guinea pigs.more importantly ,to study the effects of arsenic trioxide on CD34+,Interleukin-5,Interleukin-5Rαin the bone marrow and lung tissue.and explore the possible therapeutic mechanism of arsenic trioxide of the low dose for asthma from a bone marrow-targeting .Methods :Forty adult male guinea pigs were randomly divided into control group (group A) , asthmatic group (group B), arsenic trioxide group (group C) and dexamethasone group(group D) (ten in each group). Asthmatic model was established by ovalbumin intraperitoneal combined with inhalation . The guinea pigs were killed at 24 h after the last challenge .the total leucocytes and the petcentage of eosinophil in bronchoalveolar lavage fluid (BALF) and bone marrow (BM) were measured. The percentage of hematopoietic CD34+ cells in bone marrow were counted by flow cytometry. Immunohistochemistry detected the expression of CD34+,Interleukin - 5,Interleukin-5Rαin the BM and lung tissue. Interleukin-5 levels were measured by ELISA in BALF and BM.Results:1 The changes of total leucocytes and petcentage of eosinophils in BALF and BMThe total leucocytes and petcentage of Eos in BALF and BM of group B were significantly higher than that in groupA;compared with group B, the total leucocytes and petcentage of Eos in BALF and BM of group C and group D were decreased. the differences were significant (P < 0.05) .2 The changes of percentage of CD34+ progenitors in BMThe percentage of CD34+ progenitors in BM of group B were significantly higher than that in group A, compared with group B,it was lower in group D.but there was a remarkable increase in group C .the differences were significant (P < 0.01) .3 The changes of expression of CD34+,Interleukin-5,Interleukin-5Rαin the BM and lungThe expression of CD34+,Interleukin-5,Interleukin-5Rαin the BM and lung of group B were significantly higher than that in groupA;compared with group B,there were significant decrease in group C and group D. the differences were significant (P < 0.01) .4 The changes of Interleukin-5 levels in BALF and BM.Interleukin-5 levels in BALF and BM were significantly higher than that in group A; compared with group B,there were significant decrease in group C and group D. the differences were significant (P < 0.05) .5 Linear Correlation analysis indicated: there existed significantly positive correlation between petcentage of Eos and percentage of CD34+,Interleukin-5Rαcells in BM. the petcentage of Eos in BALF was closely correlated with expression of Interleukin-5Rαin the lung tissue. there was a strongly positive relationship of the Interleukin-5 between BALF and BM.Conclusions:1 Expression of CD34+,Interleukin-5,Interleukin-5Rαin the BM and lung tissue of guinea pig with asthma were remarkably increased.and it shows that CD34+IL-5Rαmaybe the early progenitor of Eos in BM.2 The low dose arsenic trioxide may achieve its anti-inflammatory mechanism based on reducing the production of Eos and CD34+,Interleukin-5,Interleukin-5Rαin the BM...
