Objective:By establishing one new kind of goat disuse osteoporosis model,the preventive effect of the stimulator of bone stress was researched.Materials and methods:Thirty 4-month-goats,14-15 kg body weight,were used in this study.Right hindlimbs of 20 goats were performed sciatic neurectomy to become cripple and served as unloading group and treatment group;The 10 goats were served as control group after their right sciatic nerves were exposed without resection.After operation,all goats were housed individually.The goats in treatment group were received the stress stimulation treatment 3 times every day,60min every time,the interval time is 3h.After 2-4 months treating,all the goats were given X-ray examination to observe the tibial cortical thickness and trabecular bone morphology,the tibias were removed from each goat.The tibial samples were used for biochemical,biomechanical(a three-point bending test),histological(HE staining and Masson staining) and bone mineral density testing.The statistical analysis were carried out by SPSS13.0 finally.Result:Study of X-ray have shown:compared with the control group,the tibial cortical of the unloading group became thiner,the trabecula became less and thinner;The tibial cortic- al became thicker,the tibial trabecula became more and thicker in the treatment group in comp- arison with the unloading group.Biochemical parameter:in the unloading group,the serum Ca,P and urine HOP were slightly increased,the ALP was lower(p<0.01) than those in the control group and the treatment group.There was the differences between the index of bone metabolism of the control group and the treatment group(p<0.05).BMD test:compared with the control group and the treatment group,the tibial BMD of the unloading group decreased significantly(p<0.01).There was the differences between the BMD of the control group and the treatment group(p<0.05).The BMD was showedxontrol group>treatment group>unloading group.Significant differences were observed in the biomechanical properties: The tibias of the unloading group had a lower maximum load and Elastic modulus as compared with the control group and the treatment group(p<0.01).Meanwhile,the differences was observed between the control group and the treatment group(p<0.05).It was showed: control group>treatment group>unloading group.Histological examination:HE staining showed that the tibial trabecula of the unloading group became less and thinner which was more and thicker in the control group and the treatment group.Masson staining indicated that the unloading group had less bone mineral components than the control group and the treatment group,and its bone matrix had been malmineralized.Conclusion:1,One new kind of goat disuse osteoporosis model can be established by sciatic neurotomy.2,The disuse osteoporosis can be prevented effectively by the stimulator of bone stress was researched.3,The best stress still need be further studied.
