Objective:To investigate the culture and amplifier of bone mesen- chymal stem cells in vitro and the possibility and methods of osteogenic differentiation .Method:The MSCs from marrow of C57BL mouses'femur and tibia were cultured and purified by wall adhesion in vitro.Passage 5 MSCs were chosen to induce into osteoblasts.Their morphological changes were observed by inverted phase contrast microscope.Alkaline phosphatase were detected by Ca/Co 2 weeks later and mineralization nodules were stained by von Kossa 3 weeks later.Result:The MSCs grew well with good proliferation activity in prima- ry and passage cultures. Alkaline phosphatase of osteogenic different- tiate cells significantly higher than that of control and theses cells could form mineralization nodules after exposure to osteogenic inductive medium.Conclusion:The MSCs from mouse could survive and prolifierate in vitro and could be induced into osteoblasts in special conditions and could be used as seed cells in bone-tissue engineering.
