Study On The Loss Of Heterozygosity On Chromosome 3,7,8,9,17 In Clear Cell Renal Carcinoma

Objective: Clear cell renal carcinoma (cc-RCC) is one of the most common pathological subtypes of kidney cancer. Studies show that loss of heterozygosity (LOH) on multiple genetic LOH in clear cell renal carcinoma in 10 genetic locus and to find clues for tumor suppressor genes associated with cc-RCC.Methods: Ten microsatellite polymorphism markers,including VHL-related D3S1317,D3S1038,D3S1597,FHIT-related D3S1234, D3S1300, D3S1540,Caveolin-1-related D7S522, FEZ1-related D8S261, P16-related D9S171 and P53-related TP53 on the chromosome 3,7,8,9,17.Genomic DNA LOH detection in 25 cases of cc-RCC tumor tissues and the corresponding normal tissues was performed by amplification of microsatellite DNA with PCR, polyacrylamide of gel electrophoresis and ethidium bromide staining methods.Results: Out of the 25 samples,23(92%)showed allelic loss at one or more loci on five chromosomes. The most frequent LOH 47.37%(9/19)was detected at D9S171 (P16 gene related),followed by D7S522 (34.78%,8/23, Caveolin-1 associated) , TP53 (30%,6/20, P53 associated).The rate of LOH at D3s1038,D3s1317,D3s1597 associated with VHL gene was 29.41%(5/17),25%(6/24),and 26.67%(4/15).While LOH at D3s1300,D3s1234,D3s1540 (FHIT associated) was detected in 29.41%(5/17),19.05%(4/21),and 10%(1/10).The frequency of LOH at D8S261(FEZ1 associated) was 13.01%(3/23).No significant relationship was found between LOH at twelve loci and the tumor's pathological stage (Χ~2=0.2 P =0.655 )and grade (Χ~2=4.269 P=0.092). Conclusions: LOH is a common phenomenon in cc-RCC, suggesting that genes of VHL, FHIT, Caveolin-1,P16, FZE1 and P53 may play important roles in the pathogenesis and progression of cc-RCC.
