Objective:To study the effects of the serum after taking Da Cheng Qi Tang(DCQT),emodin and magnolol on inositol 1,4,5-trisphosphate receptor (IP3R) and ryanodine receptor(RYR) expression in interstitial cells of Cajal (ICCs) in rat.Method:The ICCs in small intestine of Wistar rat were isolated, cultured and purified by adherence and flow cytometry. The cultured cells purificated in 7th day were identified by immunofluorescence and flow cytometry analysis.The ICCs were cultured with emodin,mognolol and the serum after taking DCQT, and the mRNA expression of IP3R(IP3R-1, IP3R-2,IP3R-3) and RyR(RyR-1,RyR-2,RyR-3) in ICC were detected by reverse transcription polymerase chain reaction(RT-PCR).Result:The mRNA expressions of IP3R-1,IP3R-2,IP3R-3(IP3R/β-actin) in ICC after cultureing with the serum,emodin and mognolol were significantly higher than the control ICCs(p <0.05 or p <0.01). Similarly,the mRNA expression of RyR-1,RyR-2 and RyR-3(RyR/β-actin) in ICC after cultureing with the serum,emodin and mognolol were significantly higher than the control(p<0.05 or p<0.01).Conclusion:The serum after DCQT,emodin and magnolol can in crease the content of calcium in ICC of and,significantly activate the membrance current of ICCs,and promote recovery of gastrointestinal dysfunction.
