Objective : To investigate the effects of Bletilla striata polysaccharide(BSPS) on proliferation of bone marrow hematopoietic cells and immunological function in the myelosuppressive and immuno-suppressive mice caused by cyclophosphamide.Methods : (1) The myelosuppressive and immuno-suppressive models were established in mice by intraperitopeal injection of cyclophosphamide 100mg/kg/d for 3 days. The effects of BSPS on the peripheral white blood cells(WBC),bone marrow nucleated cell( BMC) , and spleen colony-forming units ( s-CFU) , as well as the carbon clearance were studied after the various doses of BSPS were administered orally for 6 days. The effects of BSPS on the proliferation of splenocytes induced by either Con A or LPS were also determined in vitro by MTT methods.Results : (1) It was shown that the administration of BSPS 150 mg/kg and 500 mg/kg significantly elevated the peripheral WBC,BMC, and s-CHU in dose- and time-dependent manners in myelosuppressive mice caused by cyclophosphamide,compared with corresponding model groups;the effects of BSPS were similar to that of colla corii asini (CCA) when the same doses were given. (2) It was also shown that the administration of BSPS (500 mg/kg) significantly elevated the ability of macrophage engulfing carbon granula, which was equivalent to that from levamisol(e100 mg/kg)-treated group. BSPS promoted the proliferations of splenocytes induced by either Con A or LPS;the promoting effect of BSPS on the proliferation of splenocytes caused by Con A was superior to that by LPS:it significantly elevated the proliferation of splenocutes at 1μg/ml and 3μg/ml in concentration-dependent manner,but the promoting effect began declined at 10μg/ml .Conclusion : (1) BSPS can stimulate the recovery of damaged bone marrow hematopoetic function induced by cyclophosphamide , which indicate that BSPS may stimulate the proliferation of hematopoetic cells in myelosuppressive individuals . (2) BSPS may increase the functions of the non-specific immune and specific T or B cells in mice .
